      摘要 目的 探讨桃红四物汤对碘乙酸诱导的骨性关节炎模型大鼠软骨细胞自噬的影响。方法 将24只SPF级SD大鼠随机分为对照组、模型组、治疗组,每组8只。通过双侧关节腔注射碘乙酸(3mg/50μL)建立骨性关节炎模型,对照组大鼠注射等量生理盐水;造模后,治疗组大鼠使用桃红四物汤灌胃干预,模型组及对照组大鼠用等量生理盐水灌胃,持续8周。末次灌胃后检测各组大鼠压痛、热痛阈值,取膝关节制作病理切片进行SO染色,Western blot检测软骨组织内自噬相关蛋白的表达改变情况。结果 与对照组比较,模型组压痛、热刺激痛阈值下降[(267.5±60.2)g比(402.1±87.3)g,(5.3±1.1)s比(9.1±1.8)s,P<0.01],Mankin评分升高[(7.2±0.7)分比(2.1±0.4)分,P<0.01],自噬相关蛋白LC3、Beclin 1表达水平下降[(0.19±0.02)比(0.79±0.02),(0.51±0.08)比(1.18±0.05),P均<0.01],p62表达水平提高[(1.05±0.07)比(0.37±0.04),P<0.01]。与模型组比较,治疗组压痛及热刺激痛阈值提高[(359.9±66.7)g比(267.5±60.2)g,(7.2±1.5)s比(5.3±1.1)s,P均<0.01],Mankin评分下降[(4.2±0.8)比(7.2±0.7),P<0.01],自噬相关蛋白LC3、Beclin 1表达升高[(0.72±0.05)比(0.51±0.08),(0.39±0.07)比(0.19±0.02),P均<0.01],p62表达下降[(0.74±0.06)比(1.05±0.07),P<0.01]。结论 桃红四物汤可通过激活软骨细胞自噬治疗碘乙酸诱导的骨性关节炎。

      ABSTRACT Objective To investigate the effect of Taohong Siwu Decoction on chondrocyte autophagy induced by iodoacetic acid in rats with osteoarthritis Methods 24 SPF SD rats were randomly divided into control group, model group and treatment group, with 8 rats in each group. The osteoarthritis model was established by bilateral joint cavity injection of iodoacetic acid (3mg/50μL), and the control group was injected with the same amount of normal saline. After modeling, rats in treatment group were given Taohong Siwu Decoction intragastric intervention, and rats in model group and control group were given the same amount of normal saline intragastric intervention for 8 weeks. After the last intragastric administration, the threshold values of tenderness and heat pain of rats in each group were detected. Pathological sections of knee joints were taken for SO staining, and the expression changes of autophagy related proteins in cartilage tissues were detected by Western blot. Results Compared with the control group, the threshold of tenderness and heat stimulation pain in model group decreased [(267.5±60.2) g vs. (402.1±87.3) g, (5.3±1.1) s vs. (9.1±1.8) s, both P<0.01], and Mankin score increased [(7.2±0.7) points vs. (2.1±0.4) points, P<0.01], the expression levels of autophagy-related proteins LC3 and Beclin1 decreased [(0.19±0.02) vs (0.79±0.02), (0.51±0.08) vs (1.18±0.05), both P<0.01]. The expression level of P62 increased [(1.05±0.07) vs (0.37±0.04), both P<0.01]. Compared with model group, the pain threshold of tenderness and heat stimulation in treatment group increased [(359.9±66.7) g vs. (267.5±60.2) g, (7.2±1.5) s vs. (5.3±1.1) s, both P<0.01], and Mankin score decreased [(4.2±0.8) vs. (7.2±0.7), P<0.01]. The expression of autophagy related proteins LC3 and Beclin1 increased [(0.72±0.05) vs (0.51±0.08), (0.39±0.07) vs (0.19±0.02), both P<0.01], and p62 decreased [(0.74±0.06) vs (1.05±0.07), P<0.01]. Conclusion Taohong Siwu Decoction can treat iodoacetic acid-induced osteoarthritis by activating chondrocyte autophagy.
