目的 探究miR-378在乳腺癌细胞系中的表达及作用机制。方法 利用TargetScan分析miR-378的作用靶基因RAB11A。利用RT-PCR检测乳腺癌细胞系中miR-378和RAB11A的mRNA水平表达。转染MCF-7和MDA-MB-231细胞miR-378,NC(normal control)对照,采用MTT法和Transwell法检测乳腺癌细胞增殖和迁移能力的变化;采用RT-PCR和Western blot检测RAB11A的表达。荧光素酶报告分析miR-378与靶基因RAB11A之间的信号。结果 miR-378在乳腺癌细胞MCF-7,MDA-MB-231中表达上调;RAB11A在乳腺癌细胞MCF-7,MDA-MB-231中表达下调;在MCF-7,MDA-MB-231细胞中敲降miR-378后,乳腺癌细胞增殖和迁移能力与对照组相比被显著抑制,RAB11A的mRNA和蛋白表达水平明显增加,荧光素酶活性增加。结论 在乳腺癌细胞中,miR-378可通过靶向RAB11A促进乳腺癌细胞的增殖和迁移。 |

Objective To explore the expression and function of miR-378 in breast cancer cells. Methods Using TargetScan to find out how can-miR-378 act on the RAB11A gene, and Rt-PCR verified the expression of miR-378. Transfected MCf-7 and MDA-MB-231 and negative control, CCK-8 and Transwell experiment were applied to verify the proliferation and migration of breast cancer cells. The luciferase reporters were used the miR-378 binding of RAB11A. Results The expression of mir-378 was up-regulated in MCF-7 and MDA-MB-231; Rab11a was down regulated in MCF-7 and MDA-MB-231; the proliferation and migration of breast cancer cells were significantly inhibited after mir-378 was knocked down in MCF-7 and MDA-MB-231 cells, and Rab11a mRNA and protein expression levels were significantly increased, and luciferase activity was increased. Conclusion MiR-378 can promote the proliferation and migration of breast cancer cells by targeting RAB11A. |
