      Objective: To investigate the effect of Mulberry Branch Polysaccharide in regulating renal fibrosis of db/db mice based on YAP/TAZ signal pathway. Methods: Twenty four weeks’ old spontaneous db/db type 2 diabetic mice were randomly divided into model group, valsartan (20 mg/kg) group, Mulberry Branch Polysaccharide (600, 1200 mg/kg) groups with 10 mice per group. The mice were fed with high fat diet, and were intragastrically administered with drugs once a day for 60 days. The mice in normal group were fed with standard rodent food and given same volume of saline for 60 days. The urine samples were collected after 60 days’ administration. The urine volume and 24 h urine albumin (UP) were measured. And the serum creatinine (Scr) and urea Nitrogen (BUN) were determined by automatic biochemical analyzer. The fasting blood glucose (FBG) was detected after last administration with 12 h fast. Additionally, the renal samples were collected for HE staining. The renal levels of fibrosis indicators TGF-β1, fibronectin (FN), connective tissue growth factor (CTGF), collagen type I (Collagen I), inflammatory factors tumor necrosis factor (TNF-α), interleukin-6 (IL-6) and Chemokine Monocyte Chemotactic Protein-1 (MCP-1) were measured by ELISA. The expressions of Yes related protein (YAP) and TAZ in renal tissues were analyzed. Results: Compared with model group, the Scr (0.39±0.09、0.32±0.07 mg/dl) and BUN (25.75±4.75、20.62±3.68 mg/dl) levels were significantly decreased in mice treated with Mulberry Branch Polysaccharide (P<0.05). The TGF-β1 (3.76±0.52、2.53±0.38 ng/mL), FN (5.29±1.13、4.47±1.02 ng/mL), CTGF (14.52±1.22、11.24±0.87 ng/mL), CollagenⅠ (10.93±0.85、8.14±0.76 ng/mL), TNF-α (87.25±7.81、78.66±6.36 ng/L), IL-6 (34.72±3.35、21.16±2.51 ng/L) and MCP-1 (0.38±0.04、0.29±0.06 ng/L) (P<0.05). The expressions of YAP and TAZ were markedly down regulated (P<0.05). Conclusion: The protective effect of Mulberry Branch Polysaccharide on renal fibrosis of db/db mice may be associated with regulating YAP/TAZ signal pathway, reducing the formation of fibrotic factors in kidney tissue and the content of inflammatory factors, thereby playing a protective role.
