      目的 观察电针对糖尿病神经痛大鼠背根神经节磷酸化(DRG)P38丝裂原活化蛋白激酶(p-p38MAPK)表达的影响。 方法 将20只健康雄性SD大鼠随机分为空白组(n=6)、造模组(n=14),造模组大鼠采用单次腹腔注射链脲佐菌素(65mg/kg)建立糖尿病神经痛模型,造模成功的12只大鼠随机分为模型组和电针组(n=6);电针组于2周后介入电针,取双侧足三里和昆仑穴,1次/日,30 min/次,干预1周。测大鼠造模前、模后1周、模后2周、模后3周机械痛阈(PWT),采用免疫荧光法检测大鼠腰4至腰6背根神经节中p-p38MAPK阳性细胞表达。 结果 1)与空白组比较,模后1周模型组和电针组大鼠PWT无显著性差异,模后2周、模后3周模型组大鼠PWT显著降低(P<0.05);与模型组比较,模后3周电针组大鼠PWT显著升高(P<0.01);2)与空白组比较,模型组大鼠腰4至腰6背根神经节上的p-p38MAPK阳性细胞个数表达显著升高(P<0.05);与模型组比较,电针组大鼠腰4至腰6背根神经节上的p-p38MAPK阳性细胞个数表达显著降低(P<0.05)。 结论 电针对糖尿病神经痛大鼠有良好的镇痛效果,其机制可能和抑制背根神经节神经元上p-p38MAPK阳性细胞表达相关。

      Objective To observe the effects of electroacupuncture (EA) on the change of expression of Phospho-p38 mitogen activation protein kinase(p-p38MAPK) receptor in L4-L6 dorsal root ganglia (DRG) in Rats with Diabetic Neuropathic Pain (DNP). Methods Twenty healthy male SD rats were randomly divided into the Control group (n=6), Model group (n=14). Rats in Model group were induced by single streptozotocin intraperitoneal injection (65 mg/kg) to establish DNP model. Twelve diabetic neuropathic pain rats were randomly divided into DNP group (n=6) and EA group (n=6). The rats administered electroacupuncture treatment at acupoints at Zusanli and Kunlun for 30 min per day for a week. PWT was detected before streptozotocin injection, and after 1, 2, 3 weeks streptozotocin injection, and immunofluorescence was applied to observe the expression of p-p38MAPK in L4-L6 DRG. Results 1) Compared with Control group, there was no significant deference in PWT of DNP group and EA group rats, and the PWTs was significantly decreased in DNP group rats at 2 weeks and 3 weeks (P<0.05); Compared DNP group, the PWT of EA group rats was significantly increased (P<0.01); 2) Compared with Control group, the expression of p-p38MAPK in L4-L6 DRG in DNP group rats was significantly increased (P<0.05); Compared with DNP group, the expression of p-p38MAPK in L4-L6 DRG in EA group rats was significantly decreased (P<0.05). Conclusions EA has perfect analgesia for DNP. The mechanism may be related to inhibiting up-regulated expression of p-p38MAPK in DRG.
