      目的 采用紫外分光光度计法测定不同地域中药蕲蛇中多糖含量。方法 对收集不同地域中药蕲蛇,经过粉碎处理,以葡萄糖为对照品,利用苯酚-浓硫酸法显色,在检测波长490nm处,吸光度和糖浓度呈现线性关系,最终对多糖采用比色法进行定量分析。并进行精密度、重复性、稳定性及加样回收率等方法学考察。采用体外培养肝癌细胞株Bel-7404,MTT法测定蕲蛇多糖对细胞增殖的影响。结果 对照品经220-800nm进行光谱扫描,确定最大吸收波长为490nm。多糖浓度在0.000-72.240μg/mL范围内与吸光度呈现良好的线性关系,线性回归方程为:Y = 0.0111X + 0.0056。方法学考察结果显示,精密度RSD为RSD为0.18%,稳定性和重复性良好。不同地域蕲蛇多糖含量测定结果,温州、丽水、九江、三明、自贡中蕲蛇多糖含量依次为5.89%、7.017%、4.694%、8.424%、6.325%。与对照组相比,蕲蛇多糖能显著抑制肝癌细胞Bel-7404增殖,且呈剂量依赖性,高剂量组抑制率达到82.13%。结论:该方法测定不同地域蕲蛇样品,结果准确、方法简便,适用于不同地域中药蕲蛇中多糖含量的测定。制备的蕲蛇多糖对肝癌细胞株有一定抑制作用。

      Objective to determine the content of polysaccharide in Agkistrodon acutus from different regions by UV spectrophotometry. Methods Agkistrodon acutus was collected from different regions. After crushing, the glucose was used as the reference substance. The phenol concentrated sulfuric acid method was used to develop the color. At the detection wavelength of 490nm, the absorbance and sugar concentration showed a linear relationship. Finally, the polysaccharide was quantitatively analyzed by colorimetry. The precision, repeatability, stability and recovery were investigated. BEL-7404 cell line was cultured in vitro. The effect of Agkistrodon acutus polysaccharide on cell proliferation was determined by MTT method. Results the maximum absorption wavelength of the reference substance was 490 nm by scanning 220-800 nm. The linear regression equation was y = 0.0111x + 0.0056. The results of methodological study showed that the RSD of precision was 0.18%, and the stability and repeatability were good. The content of Agkistrodon acutus polysaccharide in Wenzhou, Lishui, Jiujiang, Sanming and Zigong were 5.89%, 7.017%, 4.694%, 8.424% and 6.325%, respectively. Compared with the control group, Agkistrodon acutus polysaccharide significantly inhibited the proliferation of Bel-7404 cells in a dose-dependent manner. The inhibition rate of high-dose group was 82.13%. Conclusion: the method is accurate, simple and suitable for the determination of polysaccharides in Agkistrodon acutus from different regions. The prepared Agkistrodon acutus polysaccharide has certain inhibitory effect on hepatoma cell line.
