      目的 建立一种适用于中药金钱白花蛇蛋白质含量测定的方法。方法 采用紫外分光光度法-Folin-酚试剂法测定金钱白花蛇粉末中蛋白质含量,并进行精密度、重复性、稳定性及加样回收率等方法学考察。采用体外培养肝癌细胞株SMMC7721,MTT法测定白花蛇蛋白类物质对细胞增殖的影响。结果 蛋白浓度在0-164.352 μg/mL范围内与吸光度呈现良好的线性关系,线性回归方程为:Y = 0.0036X + 0.005。方法学考察结果显示,精密度RSD为0.01%,表明仪器精密度良好。重复性RSD为0.71%,表明实验重复性良好。蛋白质平均回收率为98.953%,RSD=1.77%。与对照组相比,白花蛇蛋白各个剂量组均可抑制肝癌SMMU7721细胞增殖,且有统计学差异(P<0.05)。结论 该研究所建立的测定中药金钱白花蛇中蛋白质含量的方法简便、快速、准确,可适用于多批次、不同地区金钱白花蛇鉴别和质量控制。同时制备的金钱白花蛇蛋白类物质对肝癌细胞有一定的抑制作用。

      Objective to establish a method for the determination of protein content in Bungarus multicinctus. Methods the content of protein in Bungarus multicinctus powder was determined by ultraviolet spectrophotometry Folin phenol reagent method, and the method of precision, repeatability, stability and sample recovery were investigated. Methods: SMMC7721 cell line was cultured in vitro, and the effect of white snake protein on cell proliferation was determined by MTT method. Results there was a good linear relationship between protein concentration and absorbance in the range of 0-164.352 μ g / ml, and the linear regression equation was y = 0.0036x + 0.005. The results of methodological investigation showed that the RSD of precision was 0.01%, indicating that the precision of the instrument was good. The RSD of repeatability was 0.71%, indicating that the experimental repeatability was good. The average recovery of protein was 98.953%, RSD = 1.77%. Compared with the control group, the proliferation of smmu7721 cells in each dose group was significantly inhibited (P < 0.05). Conclusion the method is simple, rapid and accurate, which can be used for the identification and quality control of Bungarus multicinctus in different regions. At the same time, the protein of Bungarus multicinctus has certain inhibitory effect on hepatoma cells.
