      目的 探讨盐酸小檗碱对A375黑素瘤细胞增殖、凋亡和迁移的影响。方法 将黑素瘤A375细胞株根据盐酸小檗碱浓度分为4组:对照组(0μmoL/L)、低剂量组(40μmoL/L)、中剂量组(80μmoL/L)、高剂量组(120μmoL/L)。然后使用CCK8法检测各组细胞增殖情况,流式细胞仪检测细胞凋亡情况,通过Transwell法检测各组细胞的迁移能力,同时使用RT-PCR和Western blot检测A375细胞TLR4 mRNA及蛋白表达水平。采用SPSS22.0软件进行统计分析,多组间数据比较采用单因素方差分析,两组间比较采用t检验。结果 盐酸小檗碱(0、40、80、120μmoL/L)分别处理A375细胞后,随盐酸小檗碱浓度的增加,A375细胞存活率(分别为100.00±4.76、70.37±3.06、48.33±0.64、26.60±2.67)逐渐降低(F=298.78,P<0.01);细胞凋亡率逐渐升高(F=101.76,P<0.01);细胞迁移力(分别为331.67±7.64、248.67±8.08、155.33±5.03、109.33±9.02)逐渐降低(F=510.81,P<0.01);同时 TLR4 mRNA及蛋白表达量逐渐降低(p<0.05)。结论 盐酸小檗碱可以促进黑素瘤A375细胞凋亡、抑制A375细胞的增殖和迁移能力,且这种作用与盐酸小檗碱存在剂量-依赖关系。同时其抗肿瘤机制可能与抑制TLR4信号通路有关。

      Objective To explore the effect of berberine hydrochloride on proliferation, apoptosis and migration of A375 melanoma cells. Methods The A375 cell lines of malignant melanoma were divided into 4 groups according to berberine hydrochloride concentration: control group (0 moL/L), low dose group (40 moL/L), middle dose group (80 moL/L) and high dose group (120 moL/L). Cell proliferation in each group was detected by CCK8 assay, apoptosis by flow cytometry, migration ability of cells in each group was detected by Transwell assay; Meanwhile, mRNA and protein expression levels of TLR4 in A375 cells were detected by RT-PCR and Western blot. SPSS22.0 software was used for statistical analysis, one-way anova was used for data comparison between multiple groups, and t test was used for comparison between two groups. Results After treatment with berberine hydrochloride (0, 40, 80, 120 moL/L), the survival rate of A375 cells were(100.00±4.76)%,( 70.37±3.06)%, (48.33±0.64)%,( 26.60±2.67)%,respectively. decreased gradually (F=298.78,P<0.01).The apoptosis rate increased gradually (F=101.76,P<0.01). Cell migration ability (331.67±7.64, 248.67±8.08, 155.33±5.03, and 109.33±9.02, respectively) decreased gradually (F=510.81, P<0.01). At the same time, the protein and mRNA expression levels of TLR4 were significantly decreased, and these changes were dose-dependent with berberine hydrochloride.. Conclusion Berberine hydrochloride can promote the apoptosis of melanoma A375 cells and inhibit the proliferation and migration of A375 cells. Meanwhile, its anti-tumor mechanism may be related to the inhibition of TLR4 signaling pathway.
