      目的:观察单侧输尿管梗阻(UUO)小鼠的I型胶原(Col-I)及III型胶原(Col-III)的信使核糖核酸(Messenger RNA,mRNA)及蛋白的表达,探讨温阳消癥方对肾间质纤维化的影响。方法:32只C57小鼠随机分为四组:假手术组、模型组、缬沙坦组、温阳消癥组,后三组采用单侧输尿管结扎法制作肾间质纤维化模型。造模成功后缬沙坦组给予缬沙坦水悬液灌胃(浓度4.9g/mL,给药剂量16.5mg/kg);温阳消癥组给予温阳消癥方水煎剂灌胃(浓度4.9 g/mL,给药剂量49g/kg),假手术组和模型组给予等量生理盐水灌胃,治疗14天后处死各组小鼠并留取左侧肾组织标本,采用实时荧光定量多聚核苷酸链式反应(Real-time Quantitative PCR,Rt-qPCR)和蛋白免疫印迹法(Western Blot)法分别检测并分析Col-I、Col-III mRNA及蛋白表达的情况。结果:模型组Col-I、Col-III mRNA及蛋白表达均较假手术组显著上调[Col-I mRNA:(6.85±0.22)比(1.03±0.11);Col-III mRNA:(6.21±0.19)比(1.09±0.14);Col-I蛋白:(6.23±0.24)比(1.09±0.13);Col-III蛋白:(16.99±0.13)比(1.01±0.07);P<0.01];温阳消癥组及缬沙坦组Col-I、Col-III mRNA及蛋白表达均较模型组显著下调[Col-I mRNA:(1.27±0.13)比(6.85±0.22);(2.65±0.25)比(6.85±0.22);Col-III mRNA:(1.17±0.08)比(6.21±0.19);(4.11±0.14)比(6.21±0.19);Col-I 蛋白: (3.14±0.15)比(6.23±0.24);(5.18±0.22)比(6.23±0.24);Col-III 蛋白: (2.68±0.12)比(16.99±0.13);(14.14±0.19)比(16.99±0.13);P<0.01];温阳消癥组的Col-I、Col-III mRNA及蛋白表达与缬沙坦组相比均有显著下调[Col-I mRNA: (1.27±0.13)比(2.65±0.25);Col-III mRNA:(1.17±0.08)比(4.11±0.14);Col-I蛋白: (3.14±0.15)比(5.18±0.22);Col-III蛋白:(2.68±0.12)比(14.14±0.19);P<0.01]。结论:温阳消癥方可下调Col-I、Col-III mRNA及蛋白的表达,减少细胞外基质在肾间质的堆积,从而减轻UUO小鼠肾间质纤维化程度。

      Objective: To observe the expression of collagen I (Col-I) and collagen III (Col-III) mRNA and protein in mice with unilateral ureteral obstruction (UUO), and to explore the effect of Wenyang Xiaozheng Decoction on renal interstitial fibrosis. Methods: Thirty-two C57 mice were randomly divided into four groups: sham operation group, model group, valsartan group and Wenyang Xiaozheng group. The latter three groups used unilateral ureteral ligation to make renal interstitial fibrosis model. After successful modeling, valsartan group was given valsartan aqueous suspension of 0.33mg(0.2ml)/(20g mouse body weight) by gavage and Wenyang Xiaozheng group was given Wenyang Xiaozheng decoction of 0.98g(0.2ml)/(20g mouse body weight) by gavage. The sham operation group and the model group were given the same amount of normal saline by gavage. After 14 days of treatment, mice in each group were sacrificed and left kidney tissue samples were taken. The expression of Col-I and Col-III mRNA and protein were detected and analyzed by Rt-qPCR and Western Blot, respectively. Results: The expression of Col-I and Col-III mRNA and protein in the model group were significantly up-regulated compared with the sham group[Col-I mRNA: model group(6.85±0.22)vs sham group(1.03±0.11);Col-III mRNA:model group(6.21±0.19)vs sham group(1.09±0.14);Col-I protein: model group(6.23±0.24)vs sham group(1.09±0.13);Col-III protein:model group(16.99±0.13)vs sham group(1.01±0.07);(P<0.01)]; the expression of Col-I and Col-III mRNA and protein in the Wenyang Xiaozheng(WYXZ) group and the Valsartan group were significantly down-regulated compared with the model group [Col-I mRNA: WYXZ group(1.27±0.13)vs model group(6.85±0.22);Valsartan group(2.65±0.25)vs model group(6.85±0.22);Col-III mRNA: WYXZ group(1.17±0.08)比model group(6.21±0.19);Valsartan group(4.11±0.14)比vs model group(6.21±0.19);Col-I protein: WYXZ group(3.14±0.15)vs model group(6.23±0.24);Valsartan group(5.18±0.22)vs model group(6.23±0.24);Col-III protein: WYXZ group(2.68±0.12)vs model group(16.99±0.13);Valsartan group(14.14±0.19)vs model group(16.99±0.13);(P<0.01)]; the expression of Col-I and Col-III mRNA and protein in the Wenyang Xiaozheng group were significantly down-regulated compared with the Valsartan group[Col-I mRNA: WYXZ group(1.27±0.13)vs Valsartan group(2.65±0.25);Col-III mRNA:WYXZ group(1.17±0.08)vs Valsartan group(4.11±0.14);Col-I protein: WYXZ group(3.14±0.15)vs Valsartan group(5.18±0.22);Col-III protein:WYXZ group(2.68±0.12)vs Valsartan group(14.14±0.19);(P<0.01)]. Conclusion: Wenyang Xiaozheng Decoction can down-regulate the expression of Col-I, Col-III mRNA and protein, reduce the accumulation of extracellular matrix in the renal interstitium, thus reducing the degree of renal interstitial fibrosis in UUO mice.
