      目的 通过观察理气通便合剂对便秘型肠易激综合征(Constipation-predominant irritable bowel syndrome, IBS-C)模型大鼠结肠、下丘脑5-羟色胺(5-hydroxytryptamine, 5-HT)、血管活性肠肽(Vasoactive intestinal peptide, VIP)的影响,探讨理气通便合剂对IBS-C大鼠的可能作用机制。方法 将SD大鼠24只随机分为空白组,IBS-C模型组,理气通便合剂治疗组,莫沙必利治疗组,采用冰水灌胃法制备IBS-C模型。造模至7天后,予加用药物干预,中药治疗组予理气通便合剂(18.75g·kg-1·d-1)灌胃,西药组予莫沙必利水溶液(1.35mg·kg-1·d-1)灌胃,空白对照组及IBS-C模型组给予等量蒸馏水灌胃。21天后取各组大鼠下丘脑及结肠组织,酶联免疫法(ELISA)检测5-HT、VIP的水平。结果 与空白组相比,模型组大鼠结肠5-HT(35.94±2.06ng/g 比 25.88±3.10ng/g)、下丘脑5-HT(71.12±5.88ng/g 比 50.85±3.14ng/g)、结肠VIP (656.37±60.34pg/g 比 577.86±53.55pg/g)、下丘脑VIP(386.57±38.00pg/g 比 276.17±43.73pg/g)均明显升高(P<0.05),药物干预后,中药组及西药组大鼠结肠及下丘脑5-HT、VIP水平较模型组有所下降,中药组较模型组有统计学差异(P<0.05),而西药组无明显统计学差异。结论 理气通便合剂的治疗作用可能是通过抑制5-HT、VIP,调节肠道运动及分泌功能,从而达到治疗的作用。

      Abstract: Objective To Observe the effect of Liqi Tongbian Mixture on 5-HT, VIP of Colon and Hypothalamus in IBS-C model rats. Methods Twenty-four SD rats were randomly divided into control group, IBS-C model group, Liqi Tongbian mixture treatment group, and mosapride group. IBS-C model was prepared by ice-water gavage. The Chinese medicine treatment group was administered with Liqi Tongbian mixture (18.75g·kg-1·d-1), the western medicine group was administered with mosapride aqueous solution (1.35mg·kg-1·d-1), and the control group and IBS-C model group was given an equal amount of distilled water. After 21 days, the hypothalamus and colon tissues of each group were taken, and the expressions of 5-HT and VIP were detected by enzyme-linked immunoassay (ELISA). Results Compared with the control group, the levels of 5-HT in the colon and hypothalamus of the model group were increased (35.94±2.06ng/g vs 25.88±3.10ng/g, 71.12±5.88ng/g vs 50.85±3.14ng/g); meanwhile, the same trend was observed on VIP (656.37±60.34pg/g vs 577.86±53.55pg/g, 386.57±38.00pg/g vs 276.17±43.73pg/g). After the drug intervention, the levels of 5-HT and VIP in the colon and hypothalamus of the rats in the traditional Chinese medicine group and western medicine group were higher, and the Chinese medicine group has a statistical difference compared with the model group, while the western medicine group has no significant statistical difference. Conclusion The therapeutic effect of Liqi Tongbian Mixture may be achieved by inhibiting 5-HT and VIP, regulating intestinal movement and secretory function.
