目的 探讨白细胞介素6受体及其基因突变与慢性阻塞性肺疾病的相关性。方法 选择本院慢性阻塞性肺疾病(COPD)患者78例为COPD组,选择同期于我院门诊健康体检者134例作为对照组,连接酶检测技术检测IL-6R rs4845617各基因型突变型,同时检测COPD组各基因型肺功能指标。结果 与对照组比较,COPD组IL-6R rs4845617 AA基因型比例升高,IL-6R rs4845617 GG基因型比例降低(P<0.05);IL-6R rs4845617 AA基因型COPD易感性,差异有统计学意义(OR=2.418、95CI%=1.748-3.345,P <0.05)。与对照组比较,COPD组IL-6R rs4845617 A等位基因频率比例升高,IL-6R rs4845617 G等位基因频率比例降低(P<0.05);IL-6R rs4845617 A等位基因频率COPD易感性,差异有统计学意义(OR=2.911、95CI%=2.220-3.818,P<0.05)。COPD组AG、GG型FEV1、FEV1/FVC、MMEF水平高于AA型,差异有统计学意义(P<0.05);GG型FEV1、FEV1/FVC、MMEF水平高于AG型,差异有统计学意义(P<0.05)。随着慢性阻塞性肺疾病严重程度逐渐增加,AA基因型比例逐渐升高,差异有统计学意义(P<0.05)。结论 白细胞介素6受体rs4845617 基因突变型AA能增加慢性阻塞性肺疾病的易感性。 |

Objective To investigate the correlation between interleukin-6 receptor (IL-6R) and its gene mutations and chronic obstructive pulmonary disease (COPD). Methods In this study, 78 cases of COPD patients (case group) and 134 cases of health examinees (control group) in our hospital were selected. The mutants of IL-6R rs4845617 genotypes were detected by ligase assay, and the pulmonary function indexes of each genotype in COPD group were also detected. Results Compared with the control group, the proportion of IL-6R rs4845617 AA genotype increased, while that of IL-6R rs4845617 GG genotype decreased in COPD group (P < 0.05); the difference of COPD susceptibility of IL-6R rs4845617 AA genotype was statistically significant (OR=2.418、95CI%=1.748-3.345,P <0.05). Compared with the control group, the frequency of IL-6R rs4845617 A allele increased, while that of IL-6R rs4845617 G allele decreased in COPD group (P < 0.05); the susceptibility of IL-6R rs4845617 A allele to COPD was statistically significant (OR=2.911、95CI%=2.220-3.818,P<0.05). The levels of AG, GG type FEV1, FEV1/FVC, MMEF in COPD group were higher than AA type (P<0.05); the level of FEV1, FEV1/FVC and MMEF of GG type was higher than that of AG type (P<0.05). With the increase of the severity of COPD, the proportion of AA genotype increased, the difference was statistically significant(P<0.05). Conclusion The polymorphism of IL-6R (rs4845617) gene may increase the susceptibility of COPD. |
