      目的 探讨长链非编码RNA GM15621(LNCRNA GM15621)在慢性阻塞性肺病患者血清中的表达变化及其意义。方法 qPCR检测198例慢性阻塞性肺病(COPD)患者和140例健康对照者血清中长链非编码GM15621HG表达,通过酶联免疫吸附试剂盒检测血清中炎症相关因子的表达,在COPD患者来源的炎症细胞中添加不同浓度的miR-15621 mimics或PI3K(磷脂酰肌醇-3激酶)抑制剂, 处理24h后收获细胞,免疫印迹分析检测过表达LNCRNA GM15621对磷酸化AKT(蛋白激酶B)的影响。结果 COPD患者血清中LNCRNA GM15621表达显著低于正常健康对照者(P<0.05),炎症因子表达显著升高,抗炎因子水平明显降低,且LNCRNA GM15621低表达与不良预后相关,慢性阻塞性肺病患者来源的炎症细胞中添加miR-15621 mimics或PI3K抑制剂处理24h后,p-AKT表达显著降低(P<0.05),炎症因子表达显著降低,抗炎因子水平明显升高。结论 COPD患者血清中LNCRNA GM15621表达降低,LNCRNA GM15621低表达与患者不良预后相关,而过表达LNCRNA GM15621可以通过抑制PI3K/AKT信号通路抑制炎症因子的表达,上调抗炎因子水平。

      Objective To investigate the expression and significance of long-chain non-coding RNA GM15621 (LNCRNA GM15621) in the serum of patients with chronic obstructive pulmonary disease. Methods qPCR was used to detect the expression of long-chain non-coding GM15621HG in the serum of 198 patients with chronic obstructive pulmonary disease (COPD) and 140 healthy control human. Inflammatory cells were added with different concentrations of miR-15621 mimics or PI3K (phosphatidylinositol-3 kinase) inhibitors. Cells were harvested 24 hours after treatment, and the effect of overexpression of LNCRNA GM15621 on phosphorylated AKT (protein kinase B) was detected by western blot analysis. Results The expression of LNCRNA GM15621 in the serum of COPD patients was significantly lower than that of normal healthy controls (P <0.05). The expression of inflammatory factors was significantly increased and the levels of anti-inflammatory factors were significantly reduced. The low expression of LNCRNA GM15621 was associated with poor prognosis. After adding miR-15621 mimics or PI3K inhibitors to the inflammatory cells for 24 hours, the expression of p-AKT was significantly reduced (P <0.05), the expression of inflammatory factors was significantly reduced, and the levels of anti-inflammatory factors were significantly increased. Conclusion The expression of LNCRNA GM15621 in serum of patients with COPD is reduced, and the low expression of LNCRNA GM15621 is associated with poor prognosis of patients. Overexpression of LNCRNA GM15621 can inhibit the expression of inflammatory factors and up-regulate the levels of anti-inflammatory factors by inhibiting the PI3K/AKT signaling pathway.#$NLKeywords: long non-coding RNA; chronic obstructive pulmonary disease; inflammatory factors; phosphatidylinositol-3 kinase; inhibitor
