      目的 探讨芹菜素对大鼠烧伤创面微循环的改善作用并研究其机制。方法 采用压力0.03MPa、106℃持续5s的方法制备深II度烧伤大鼠模型;设假烧伤组(0.9%氯化钠溶液),模型组(0.9%氯化钠溶液),磺胺嘧啶银软膏单药组,磺胺嘧啶银软膏+芹菜素低、中、高剂量组(12.5、25、50 mg/kg),各30只,疗程14d。观察各组大鼠的创面大体情况、HE染色法行创面组织病理学检查,计算烧伤创面愈合率,检测创面组织微循环血流灌注值(MPD)、微血管密度(MVD)和创面组织含水量,Western blot法检测表皮生长因子(EGF)、碱性成纤维细胞因子(b-FGF)、血管内皮生长因子(VEGF)蛋白表达。结果 与模型组比较:磺胺嘧啶银软膏+芹菜素低、中、高剂量组和磺胺嘧啶银软膏单药组烧伤大鼠创面新生上皮生长活跃、病理性形态结构改变明显改善,创面愈合率升高;MPD、MVD升高而创面组织含水量降低,EGF、b-FGF、VEGF蛋白表达量升高,差异均具有统计学意义(P<0.05或P<0.01)。与磺胺嘧啶银软膏单药组比较:磺胺嘧啶银软膏+芹菜素高剂量创面组织病理性形态结构改变改善更为明显,创面愈合率升高,MPD、MVD升高且创面组织含水量降低,EGF、b-FGF、VEGF蛋白表达量升高,差异具有统计学意义(P<0.05或P<0.01)。结论 芹菜素能够通过改善大鼠深II度烧伤创面微循环而促进创面愈合,其机制可能与促进血管新生有关。
英文摘要:
      Objective To investigate the effect of Apigenin on the microcirculation of burn wounds in rats and study its mechanism. Methods The model rats with deep II degree burn injury were made by using a pressure of 0.03MPa and 106 ℃ for 5 seconds; setting sham burn group (saline), model control group (saline), Sulfadiazine silver ointment group, Sulfadiazine silver+ Apigenin low-, medium- and high-dose groups (12.5, 25, 50 g/kg), n=30, the drugs were given for 14d. The wound status were observed and the burn wound healing rate were calculated; the histopathological examination of the wound by HE staining; the microcirculation blood flow perfusion value (MPD), microvascular density (MVD) and water content in wound tissue were detected, the expression of EGF, bFGF, VEGF proteins were determined by Western blot method. Results Compared with the model group, the neonatal epithelium of burned rats grew actively in Sulfadiazine silver+Apigenin low-, medium- and high-dose groups and Sulfadiazine silver ointment group, the pathological structure changes were improved, the wound healing rate were increased; the MPD, MVD were increased and the water content were decreased; the expression of EGF, bFGF, VEGF were increased, all of the the differences were significant (P<0.05 or P<0.01). Compared with Sulfadiazine silver ointment group, the pathological morphological structure of the wound tissue was more improved of the rats in Sulfadiazine silver+Apigenin high-dose group, the wound healing rate was increased, the MPD, MVD were increased and the water content was decreased, the expression of EGF, bFGF, VEGF were increased, all of the the differences were significant (P<0.05 or P<0.01). Conclusion Apigenin can promote wound healing by improving the microcirculation of deep second-degree burn wounds in rats, and its mechanism may be related to promoting angiogenesis.
