      目的 观察茄根醇提物对Ⅱ度冻伤SD大鼠表观皮损修复及病理形态、血液流变学的干预作用。方法 将15只SD大鼠随机分为生理盐水、茄根醇提物、尿素维E乳膏组各5只,液氮法造模,每日两次1mL/100g规格擦涂给药,观察表观皮肤损伤修复、镜下病理形态、血液流变学指标。结果 示茄根醇提组、尿素维E乳膏组造模第6天表观指标较生理盐水组显著好转[(-2.20±0.837)、(-2.80±1.304)比(-0.60±0.548),P<0.05],茄根醇提组与尿素维E乳膏组差异无统计学意义(P>0.05)。病理形态示造模第18天茄根醇提组、尿素维E乳膏组镜下炎细胞较少、已有肉芽组织形成。造模第18天尿素维E乳膏组血液流变学指标血浆黏度[(1.42±0.08)mPa.S比(1.34±0.06)mPa.S,P<0.05]、纤维蛋白原[(3.14±0.17)g/L比(2.94±0.12)g/L,P<0.05]高于生理盐水组。茄根醇提组较生理盐水组稍有降低但差异无统计学意义(P>0.05)。茄根醇提组全血黏度切变率1.00/s、5.00/s、30.00/s、200.00/s[(30.64±7.79)mPa.S比(53.27±16.83)mPa.S、(13.10±2.80)mPa.S比(19.76±4.85)mPa.S、(7.30±1.29)mPa.S比(9.51±1.55)mPa.S、(5.36±0.81)mPa.S比(6.31±0.64)mPa.S,P<0.05]、血浆黏度[(1.34±0.02)mPa.S比(1.42±0.08)mPa.S,P<0.05]、红细胞聚集指数[(5.64±0.68)比(8.30±1.94),P<0.05]、纤维蛋白原[(2.95±0.05)g/L比(3.14±0.17)g/L,P<0.05]均显著低于尿素维E乳膏组。结论 茄根醇提物能促进Ⅱ度冻伤SD模型大鼠皮肤修复,促进愈合,作用机制可能与抗炎作用有关。

      Objective To investigate the effect of ethanol extraction components from eggplant root on apparent skin lesion repair, pathomorphology and hemorheology in SD rats with second degree frostbite. Methods Fifteen SD rats were randomly divided into three groups: normal saline group (n=5), eggplant root ethanol extraction group (n=5) , Urea and Vitamin E Cream group (n=5) and smeared with 1mL/100g in each group twice a day. Made the second degree frostbite model by liquid nitrogen method. Observed the repair of apparent skin damage, pathological morphology and hemorheological indexes. Results The apparent index showed that after 6 days of modeling, the difference of apparent index score in eggplant root ethanol extraction group and Urea and Vitamin E Cream group was significantly better than that in normal saline group [(-2.20±0.837)、(-2.80±1.304) vs (-0.60±0.548),all P<0.05], but there was no significant difference between eggplant root ethanol extraction group and Urea and Vitamin E Cream group(P>0.05). Compared to the controls, pathomorphology index showed that 18 days after molding there were few inflammatory cells and granulation tissue formed in eggplant root ethanol extraction group and Urea and Vitamin E Cream group. Hemorheology index showed that 18 days after modeling the plasma viscosity [(1.42±0.08)mPa.S vs (1.34±0.06)mPa.S,P<0.05]and fibrinogen [(3.14±0.17)g/L vs (2.94±0.12)g/L,P<0.05]in the Urea and Vitamin E cream group were higher than those in the normal saline group. The ethanol extraction group were slightly lower than those in the normal saline group, but there was no significant difference between the two groups (P> 0.05). At the same time, the whole blood viscosity at the shear rate of 1.00/s, 5.00/s, 30.00/s, 200.00/s of the ethanol extraction group were significantly lower than those in the Urea and Vitamin E cream group [(30.64±7.79)mPa.S vs (53.27±16.83)mPa.S、(13.10±2.80)mPa.S vs (19.76±4.85)mPa.S、(7.30±1.29)mPa.S vs (9.51±1.55)mPa.S、(5.36±0.81)mPa.S vs (6.31±0.64)mPa.S,all P<0.05]. The plasma viscosity[(1.34±0.02)mPa.S vs (1.42±0.08)mPa.S,P<0.05], erythrocyte aggregation index[(5.64±0.68) vs (8.30±1.94),P<0.05], fibrinogen[(2.95±0.05)g/L vs (3.14±0.17)g/L,P<0.05] of the ethanol extraction group were significantly lower than those in the Urea and Vitamin E cream group. Conclusion For mild frostbite, there is no persistent endothelial injury and bleeding in local tissue, and the key point of treatment is anti-inflammation. Ethanol extraction of eggplant root can improve the repair of frostbite skin and effectively promote healing, its mechanism may be related to anti-inflammatory effect.
