目的 通过观察姜黄素对血管内皮生长因子(VEGF)刺激干预后的HaCaT细胞增殖及细胞内微小RNA-203(miR-203)表达的影响,探讨姜黄素治疗银屑病目的的可能作用机制。方法 使用CCK-8法分别检测不同浓度姜黄素(0、19.5、39、78.1、156.2、312.5、625、1250、2500、5000ng/ml)对25ng/ml VEGF刺激干预后的HaCaT细胞增殖的影响。通过实时荧光定量PCR法(Real-time PCR法)检测25ng/ml VEGF、1380ng/ml姜黄素、5000ng/ml姜黄素分别及联合作用下对HaCaT细胞内miR-203表达情况的影响。结果 姜黄素在实验浓度范围内呈浓度依赖性抑制HaCaT细胞增殖,2500ng/ml姜黄素明显抑制细胞增殖。与空白组比较,miR-203在25ng/ml VEGF诱导下的HaCaT细胞中表达明显上调(P<0.05),而5000ng/ml姜黄素能明显抑制HaCaT细胞中miR-203的表达(P<0.05),同样能下调25ng/ml VEGF刺激干预下HaCaT细胞中miRNA-203的表达(P<0.05)。结论 抑制角质形成细胞的过度增殖是姜黄素治疗银屑病的作用机制之一,上述作用机制可能是姜黄素通过下调细胞内miR-203的表达来实现的。 |

Objective To explore the possible mechanism of curcumin in the treatment of psoriasis by observing the effect of curcumin and vascular endothelial growth factor (VEGF) on cell proliferation and microRNA-203(miR-203) expression of HaCaT cell. Methods Using CCK8 assay to detect the influence of curcumin with different concentrations(0、19.5、39、78.1、156.2、312.5、625、1250、2500、5000ng/ml) on the proliferation of HaCaT cells induced by 25ng/ml VEGF;25ng/ml VEGF, 1380ng/ml curcumin and 5000ng/ml curcumin were treated cells either independently or together, then real-time PCR assay was used to detect the expression of miR-203 on the cells. Result Curcumin inhibited the proliferation of HaCaT cells in a concentration-dependent manner in the range of 0~5000ng/ml. 2500ng/ml curcumin inhibited the cell proliferation significantly(P<0.05). Compared with the blank control group, 25ng/ml VEGF promoted the expression of miR-203 in HaCaT cells (t=17.47,P =0.0033), while 5000ng/ml curcumin significantly inhibited the expression(t=17.83,P =0.0031) and decreased the effect of 25ng/ml VEGF(t=22.49,P =0.002). Conclusion One of the mechanisms in the treatment of psoriasis with curcumin is inhibiting excessive proliferation of keratinocytes. The mechanism maybe achieved by the decrease of miR-203 expression in cells. |
