      目的:探讨鼠尾草酚对脓毒症大鼠肺损伤的保护作用及转化受体电位阳离子通道亚家族M成员6(TRPM6)、TRPM7表达的影响。方法:用LPS制备脓毒症大鼠肺损伤模型,造模1h后鼠尾草酚低、中、高剂量组分别股静脉给予1ml鼠尾草酚,剂量依次为5mg/kg、10mg/kg、20mg/kg,对照组和模型组分别股静脉给予1ml生理盐水,24 h后检测PaO2、PaCO2、PaO2/FiO2,计算W/D、对肺组织进行病理学检查,检测肺组织中MPO和SOD活性及TRPM6和TRPM7 mRNA表达水平,检测支气管肺泡灌洗液中蛋白、IL-6和TNF-ɑ含量。结果:对照组肺泡结构清楚,无炎性细胞浸润痕迹;模型组肺间室增大,肺间质炎症细胞浸润,肺间隔发生断裂,肺毛细血管充血伴血外;各鼠尾草酚剂量组肺损伤程度较模型组明显减轻,且呈剂量依赖性。与对照组比较,模型组和各鼠尾草酚剂量组大鼠PaO2、PaO2/FiO2和SOD降低,PaCO2、W/D、MPO、蛋白、IL-6、TNF-ɑ、TRPM6和TRPM7 mRNA水平升高(P<0.05);与模型组相比,各鼠尾草酚剂量组大鼠PaO2、PaO2/FiO2和SOD升高,PaCO2、W/D、MPO、蛋白、IL-6、TNF-ɑ、TRPM6和TRPM7 mRNA水平降低,且呈剂量依赖性(P<0.05)。结论:鼠尾草酚可能通过降低TRPM6和TRPM7的表达水平,对脓毒症大鼠肺损伤具有保护作用。

      To investigate the protective effect of salvianol on lung injury in sepsis rats and its effect on the expression of TRPM6 and TRPM7. Methods: The lung injury model in sepsis rats was made by LPS. After 1 hour, the low, middle and high dose groups of salvianol were given 1 ml salvianol via femoral vein, and the dosage was 5 mg/kg, 10 mg/kg and 20 mg/kg respectively. The control group and model group were given 1 ml saline via femoral vein respectively. PaO2, PaCO2 and PaO2/FiO2 were detected 24 hours later. W/D was calculated and pathology of lung tissue was performed. The activity of MPO and SOD, the expression of TRPM6 and TRPM7 in lung tissue and the contents of protein, IL-6 and TNF-ɑ in bronchoalveolar lavage fluid were detected. Results: The alveolar structure of the control group was clear and there was no infiltration of inflammatory cells; the model group had enlarged pulmonary compartment, infiltration of inflammatory cells in pulmonary interstitium, rupture of pulmonary septum, pulmonary capillary congestion with blood; the degree of lung injury in each group of salvianol dosage was significantly alleviated, and it is dose-dependent. Compared with the control group, the PaO2, PaO2/FiO2 and SOD in the model group and the salvianol dosage group were significantly decreased, while the levels of PaCO2, W/D, MPO, protein, IL-6, TNF-ɑ, TRPM6 and TRPM7 were significantly increased(P<0.05). Compared with model group, the PaO2, PaO2/FiO and SOD in the salvianol dosage group were significantly increased, while the levels of PaCO2, W/D, MPO, protein, IL-6, TNF-ɑ TRPM6 and TRPM7 were significantly decreased in a dose-dependent manner(P<0.05). Conclusion: Salvianol may have protective effect on lung injury in sepsis rats by reducing the expression levels of TRPM6 and TRPM7.
