      目的:探索姜黄素逆转人结肠癌耐药细胞株HCT-116/5FU作用及机制。方法:姜黄素、5-FU诱导细胞凋亡,采用MTT法检测细胞增殖以明确最佳实验条件;AnnexinV-FITC试剂盒检测细胞凋亡率;Western-blot分析耐药细胞株内cleaved-Caspase3、IL-6、STAT3蛋白表达水平。结果:姜黄素能逆转人结肠癌耐药细胞株HCT-116/5FU的耐药程度;对照组、5-FU组、姜黄素组、联合用药组凋亡率分别为(2.5±2.1)%、(6.5±3.2)%、(19.9±7.2)%和(56.0±12.1)%;耐药细胞株中IL-6、STAT3蛋白表达明显高于亲本株;姜黄素的使用降低了耐药细胞株中IL-6、STAT3的蛋白表达量。结论:姜黄素能逆转人结肠癌耐药细胞株HCT-116/5FU的耐药,其机制可能与下调IL-6/ STAT3有关。

      Objective: To explore the effect and mechanism of the drug-resist reversal caused by curcumin on human colon cancer cell line Hct-116/5FU. Methods: Curcumin and 5-FU was used to induce apoptosis; MTT assay was used to determine cell proliferation and determine the experimental conditions based on the previous results. AnnexinV-FITC kit was used to detect the apoptosis rate. Western-blot was used to evaluate the expression levels of cleaved-Caspase3, IL-6 and STAT3 in drug-resistant cell line. Results: Curcumin can reverse the resistance of human colon cancer cell line HCT-116/5FU; The apoptosis rates of the control group, 5-FU group, curcumin group, and combination drug group were (2.5 ± 2.1)%, (6.5 ± 3.2)%, (19.9 ± 7.2)%, and (56.0 ± 12.1)%, respectively; The expression of IL-6 and STAT3 protein in drug-resistant cell lines was significantly higher than the parental strains; Curcumin reduced the protein expression levels of IL-6 and STAT3. Conclusion: Curcumin can reverse the resistance of human colon cancer resistant cell line HCT-116/5FU, and its mechanism may be related to the down-regulation of IL-6 / STAT3.
