      目的 探讨山茱萸提取物对乳腺癌细胞MDA-MB-231侵袭和转移的抑制作用及机制。方法 设MDA-MB-231细胞组、顺铂组(100.0μg/ml)、山茱萸提取物低剂量组(100.0μg/ml)、高剂量组(200.0μg/ml);以上各组每孔设6个平行样,培养72h。培养结束后,MTT法测定细胞活力,结晶紫染色测定单克隆形成数目,刮擦伤口愈合试验测定细胞迁移,transwell室测定细胞侵袭,RT-PCR法及WESTERN-BLOT法测定IGF-1R、Akt基因和蛋白水平。结果 与MDA-MB-231细胞组比较,顺铂组、山茱萸提取物低、高剂量组OD值、存活率、单克隆形成数目、迁移率、穿膜数、IGF-1R、Akt mRNA和蛋白表达降低(P<0.05);与顺铂组比较,山茱萸提取物低剂量组OD值、存活率、单克隆形成数目、迁移率、穿膜数、IGF-1R、Akt mRNA和蛋白表达升高(P<0.05),山茱萸提取物高剂量组OD值、存活率、单克隆形成数目、迁移率、穿膜数、IGF-1R、Akt mRNA和蛋白表达降低(P<0.05);与山茱萸提取物低剂量组比较,山茱萸提取物高剂量组OD值、存活率、单克隆形成数目、迁移率、穿膜数、IGF-1R、Akt mRNA和蛋白表达降低(P<0.05)。结论 山茱萸提取物对乳腺癌细胞MDA-MB-231增殖、侵袭、转移具有明显抑制作用;其机制可能与山茱萸提取物抑制IGF-1R、Akt mRNA和蛋白的表达进而抑制IGF-1R/Akt信号通路的激活有关。

      Objective: To investigate the inhibitory effect and mechanism of Cornus officinalis extract on invasion and metastasis of breast cancer cell MDA-MB-231. Methods: MDA-MB-231 cell group, cisplatin group (100.0μg/ml), Cornus extract low-dose group (100.0μg/ml), Cornus extract high-dose group (200.0μg/ml) were established, and six parallel samples in each hole of the above groups were cultured for 72h. At the end of culture, MTT method was used to measure cell viability, and crystal violet staining was used to measure the number of monoclonal formation, scraping wound healing test used to measure cell migration, Transwell chamber used to measure cell invasion, RT-PCR and Western-blot used to measure IGF-1R, Akt gene and protein levels. Results: Compared with MDA-MB-231 cell group, the OD value, survival rate, number of monoclonal formation, migration rate, number of membrane penetration, the IGF-1R, Akt mRNA and protein expression were significantly decreased in cisplatin group and Cornus extract low-dose and high-dose group (P<0.05). Compared with cisplatin group, the OD value, survival rate, number of monoclonal formation, migration rate, number of membrane penetration, the IGF-1R, Akt mRNA and protein expression were significantly increased in Cornus extract low-dose group (P<0.05). And the OD value, survival rate, number of monoclonal formation, migration rate, number of membrane penetration, the IGF-1R, Akt mRNA and protein expression in Cornus extract high-dose group significantly decreased(P<0.05). Compared with the Cornus extract low-dose group, the OD value, survival rate, number of monoclonal formation, migration rate, number of membrane penetration, the IGF-1R, Akt mRNA and protein expression in the Cornus extract high-dose group were significantly decreased(P<0.05). Conclusion: The Cornus officinalis extract can inhibit the proliferation, invasion and metastasis of MDA-MB-231 cells, and its mechanism may be related to the inhibition of IGF-1R, Akt mRNA and protein expression and then the inhibition of IGF-1R/Akt signal pathway activation.
