      目的 碘乙酸(Monoiodoacetic acid, MIA)注射是建立骨性关节炎(Osteoarthritis, OA)动物模型的经典方法,本研究通过大鼠双侧膝关节腔注射不同浓度MIA建立大鼠OA模型,探究不同浓度碘乙酸不同处理时间致大鼠膝骨性关节炎的模型规律。方法 将72只200g左右的清洁级雄性SD大鼠随机分为4组,对照组、低剂量组、中剂量组、高剂量组每组各18只。对照组双侧膝关节各注射50μl生理盐水,低剂量组每侧膝关节注射1 mg/50μl的MIA溶液(超纯水配置)50μl,中剂量组每侧注射3 mg/50μl的MIA溶液50μl,高剂量组每侧注射6 mg/50μl的MIA溶液50μl。比较注射后第1天,第1、2、3、4、5周运动功能、疼痛学改变、关节软骨病理变化及Mankind评分。结果:各实验组在造模两天后就出现轻微OA症状,造模后3周出现典型的病理、疼痛学改变,具有剂量效应和快速建模的特点。结论采取关节腔注射MIA能快速建立大鼠OA模型,并且具有剂量效应,随着剂量的加大,大鼠关节损伤越严重,在第3周至第4周时所表现骨性关节炎症状最为明显,是研究早期骨关节炎的理想模型。

      Objectives Monoiodoacetic acid (MIA) injection is the classic method to establish an animal model of osteoarthritis(OA). In this study, rat models of OA were established by injecting different concentrations of MIA into bilateral knee joint cavity to study on the regularity of rat knee OA model with different concentration of MIA and different treatment time. Method 72 clean male SD rats of about 200g were randomly divided into 4 groups, control group and the low dose group, the middle dose group and the high dose group were 18 in each group. In the control group, 50μl normal saline was injected into the two sides of the knee joint. In the low dose group, each side of the knee joint was injected with 1 mg/50μl MIA solution (ultra pure water configuration) 50μl, the medium dose group was injected with 3 mg/50μl MIA solution 50μl, and the high dose group was injected with 6 mg/50μl MIA solution 50μl. Motor function, pain changes, pathological changes of articular cartilage and Mankind scores were compared on the first day, 1,2,3,4,5 weeks after injection. Result The experimental group showed mild OA symptoms two days after modeling, and 3 weeks after modeling, there were typical pathological and pain changes, which had the characteristics of dose effect and rapid modeling. Conclusion Take the intra-articular injection of MIA can quickly establish a rat model of OA, and has the dose effect, with the increasing doses, the rats joint injury is more serious, in third to fourth weeks when the symptoms of osteoarthritis is the most obvious, is an ideal model for studying early osteoarthritis
