      目的 探究芍药醇对脓毒症大鼠急性肺损伤的保护作用及其机制。方法 将18只大鼠分成对照组,脂多糖(LPS)诱导的脓毒症模型组,芍药醇治疗组,每组6只。LPS处理24h后,氧化应激试剂盒检测肺组织总抗氧化能力(T-AOC)、丙二醛(MDA)水平和过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性,酶联免疫吸附实验(ELISA)检测肺组织肿瘤坏死因子α(TNF-α)、白细胞介素 6(IL-6)、转化生长因子-β(TGF-β)水平,Western blot检测肺组织炎症、氧化应激及凋亡相关蛋白表达,实时定量PCR(QRT-PCR)检测相关miRNA表达。结果 相比于对照组,模型组MDA 水平显著提高,T-AOC 水平显著下降,CAT和SOD活性减低;TNF-α、IL-6、TGF-β水平升高;高迁移率蛋白1(HMGB1)、半胱天冬氨酸酶9剪切体(cleaved-Caspase9)表达增多,B淋巴细胞瘤-2(Bcl-2)、核转录因子2(Nrf2)表达减少;非编码RNA miR-126、miR-223水平降低,miR-21水平升高。芍药醇处理脓毒症大鼠,显著升高T-AOC 水平,抑制MDA 水平升高,CAT和SOD活性增强;TNF-α、IL-6、TGF-β水平降低;HMGB1、cleaved-Caspase9蛋白表达减少,Bcl-2、Nrf2蛋白表达增多;miR-126、miR-223水平升高,miR-21水平降低。结论 芍药醇抑制脓毒症大鼠炎症反应、氧化应激及细胞凋亡,对急性肺损伤具有保护作用。

      Objective To observe the protective effect and mechanism of paeonol on acute lung injury in septic rats. Methods 18 rats were divided into control group (n=6), lipopolysaccharide (LPS)-induced sepsis model group (n=6) and paeonol group (n=6). After 24 hours of LPS treatment, the levels of total antioxidant capacity (T-AOC), malondialdehyde (MDA), catalase (CAT) and superoxide dismutase (SOD) activity were detected. The concentrations of tumor necrosis factor alpha (TNF-α), interleukin-6 (IL-6) and transforming growth factor-β (TGF-β) were detected by immunosorbent assay (ELISA). The expressions of HMGB1, Caspase9, cleaved-Caspase9, Bcl-2 and Nrf2 were detected by Western blot. The levels of miR-126, miR-223 and miR-21 were detected by real-time quantitative PCR (QRT-PCR). Results MDA, TNF-α, IL-6, TGF-β, HMGB1, cleaved-Caspase9 and miR-21 level of the lung tissue in sepsis group were significantly increased compared with control group, while these indices were significantly decreased in paeonol group compared with sepsis group. T-AOC, CAT, SOD, miR-223, miR-126, Nrf2 and Bcl-2 level of the lung tissue in sepsis group were significantly decreased compared with control group, while these indices were significantly increased in paeonol group compared with sepsis group. Conclusion Paeonol inhibits inflammatory response, oxidative stress and apoptosis in septic rats, and it has protective effects on acute lung injury.
