      目的:观察针刺“百会”透“曲鬓”穴对脑出血(ICH)大鼠自噬相关蛋白的影响,探究针刺促进ICH后大鼠神经功能恢复的作用机制。方法:将80只雄性SD大鼠(250-300 g)随机分成4组,假手术组、模型组(50 μL自体血注入尾状核)、非穴位组(平行中线距“百会”穴1 cm 处)、针刺组(患侧“百会”透“曲鬓”),每个亚组20只。于行为学评分(前肢放置试验、拐角试验)结束后取患侧出血半暗带区脑组织,用免疫组化检测p62蛋白表达,用Western blot检测p62、Beclin-1表达含量变化。结果:在ICH后7天模型组出现明显神经功能缺损体征(均P<0.01,转角试验(%):28±8.37 vs. 60±15.81;前肢放置试验(%):36±8.94 vs. 88±8.37),并上调脑组织p62、Beclin-1蛋白表达含量(均P<0.01,p62:0.85±0.07 vs. 0.37±0.03;Beclin-1:0.57±0.05 vs. 0.28±0.04);而针刺干预能够明显提高ICH大鼠行为学评分(均P<0.01 vs. 模型组/非穴位组,44±11.41 vs. 28±8.37 or 22±8.37; 64±11.40 vs. 36±8.94 or 38±8.37),并下调ICH大鼠脑组织p62,上调Beclin-1蛋白表达含量(均P<0.01 vs. 模型组/非穴位组,0.61±0.05 vs. 0.85±0.07 or 0.82±0.06; 0.82±0.07 vs. 0.57±0.05 or 0.61±0.08)。结论:针刺能下调ICH大鼠脑组织p62蛋白表达,上调Beclin-1蛋白表达,进而改善大鼠神经功能缺损体征。

      Objective To observe the effect of acupuncture on autophagy related proteins in rats with intracerebral hemorrhage (ICH) and explore the mechanism of acupuncture promoting the recovery of neurological function in ICH rats. Methods Eighty male SD rats were randomly divided into 4 groups. Sham operation group (n = 20), model group (50 μ L autologous blood injected into caudate nucleus, n = 20), non-acupoint group (parallel midline distance from Baihui point 1 cm, n = 20), acupuncture group (affected side “Baihui” through “Qubin”, n = 20). Neurological deficit in the various groups were assessed based on forelimb placement test and corner test. the expression of p62 protein was detected by immunohistochemistry, and the expression of p62 and Beclin-1 was detected by Western blot. Results The rats had more serious brain injury on the 7th day in the model group (P <0.01, corner test(%):28±8.37 vs. 60±15.81;forelimb placement test(%):36±8.94 vs. 88±8.37), and the expression of p62 and Beclin-1 protein in brain tissue was up-regulated (all P < 0.01, p62:0.85±0.07 vs. 0.37±0.03;Beclin-1:0.57±0.05 vs. 0.28±0.04). Acupuncture intervention could significantly improve the behavioral score of ICH rats (all P < 0.01vs. model group or non-acupoint group, 44±11.41 vs. 28±8.37 or 22±8.37; 64±11.40 vs. 36±8.94 or 38±8.37), and down-regulated p62 and up-regulated the expression of Beclin-1 protein in the brain tissue of ICH rats (all P < 0.01vs. model group or non-acupoint group, 0.61±0.05 vs. 0.85±0.07 or 0.82±0.06; 0.82±0.07 vs. 0.57±0.05 or 0.61±0.08). Conclusions acupuncture can down-regulate the expression of p62 protein and up-regulate the expression of Beclin-1 protein in brain tissue of ICH rats, to improve neurological deficit in ICH rats.
