| 目的 检测结肠癌(Colorectal carcinoma, CRC)中核因子E2相关因子2(Nrf2)和Kelch样环氧氯丙胺相关蛋白1(Keap1)的表达,分析二者与增殖细胞核抗原(PCNA)的关系。方法 本实验以96例CRC术后的肿瘤组织作为观察组,选择距肿物边缘>3厘米正常结肠粘膜组织作为对照组。应用免疫组化法、Western Blot法检测二组中Nrf2、Keap1蛋白的表达,应用荧光实时定量PCR法检测二组中Nrf2、Keap1 mRNA的表达。应用相关分析观察Nrf2与PCNA、Keap1与PCNA的关系。结果 免疫组化方法显示二组中Nrf2和Keap1蛋白的阳性率差别有统计学意义(P<0.05)。观察组中Nrf2和Keap1的阳性率在不同肿物最大径、浸润深度、淋巴结转移、远处转移和TNM分期分组中的表达差别有统计学意义(P<0.05),Nrf2和Keap1的阳性率在不同性别、年龄和分化程度分组中的表达差别无统计学意义(P>0.05),观察组中Nrf2和Keap1的表达与预后有关(P<0.05)。观察组中Nrf2和PCNA之间具有正相关性(P<0.05),Keap1和PCNA之间具有负相关性(P<0.05)。Western Blot法显示二组中Nrf2和Keap1的半定量表达差别有统计学意义(P<0.05)。荧光PCR法显示二组中Nrf2和Keap1 mRNA的表达差别有统计学意义(P<0.05)。结论 Nrf2高表达、Keap1低表达在CRC的形成和进展有重要作用。Nrf2/Keap1的协同作用可能通过对细胞增殖的调节起作用,术后检测Nrf2和Keap1的表达对判断预后有一定价值。 |

| Objective To detect expressions of Nrf2 and Keap1 in colorectal carcinoma, analyze theri relationship with PCNA index. Methods 96 cases of colorectal carcinoma were selected as observation group, normal colorectal mucosa tissues(> 3cm from tumor edge) were selected as control group. Expressions of Nrf2 and Keap1 were detected by IHC, western Blot and fluorescence PCR in two groups. Relationship between Nrf2 and PCNA, Keap1 and PCNA proliferation index were analyzed in observation group. Results IHC results: Expression of Nrf2 and Keap1 were significantly different in two groups. Positive rate of Nrf2 and Keap1 were significantly different in different maximum diameter, infiltration depth, lymph node metastasis, distant metastasis and TNM staging in observation groups. Positive rate of Nrf2 and Keap1 were not significant difference in different sex, age and differentiation degree. The expressions of Nrf2 and Keap1 were related to prognosis in observation group. Expression of Nrf2 has a Positive correlation with PCNA, Keap1 has a negative correlation with PCNA in observation group. Semi-quantitative expressions of Nrf2 and Keap1 were statistically significant by Western Blot method in two groups. Expressions of Nrf2 and Keap1 mRNA were statistically significant by fluorescence PCR in two groups. Conclusion The higher Nrf2 expression, lower Keap1 can promote formation and progression of colorectal carcinoma. Nrf2/Keap1 may play a role in regulate cell proliferation. Expressions of Nrf2 and Keap1 have certain value to judge prognosis in colorectal carcinoma. |
