      目的 探讨lncRNA TUG1调控脓毒症炎症反应的机制。方法 选取脓毒症患者(脓毒症组)及健康者(对照组)各30例,实时定量PCR检测血清lncRNA TUG1和HMGB1表达水平,ELISA检测炎症因子IFN-γ、IL-10、TNF-α和IL-6浓度。过表达TUG1慢病毒感染细菌脂多糖(LPS)处理的THP-1细胞,MTT检测细胞活力改变,western blot检测HMGB1表达,ELISA检测巨噬细胞炎性因子浓度。结果 相比于对照组,脓毒症患者外周血lncRNA TUG1水平显著降低,HMGB1蛋白表达增多,两者呈负相关(P<0.01),促炎因子IFN-γ、TNF-α和IL-6浓度显著升高,抗炎因子IL-10水平明显下降;数据库starBase预测TUG1调控靶基因HMGB1,LPS处理的THP-1细胞过表达TUG1后,细胞活力增强,HMGB1表达减少,炎性因子IL-6和TNF-α水平降低。结论 lncRNA TUG1抑制脓毒症炎症反应,为脓毒症诊断及治疗靶点提供科学依据。
英文摘要:
      Objective To investigate the mechanism of lncRNA TUG1 in regulating the inflammatory response of sepsis. Methods 30 patients with sepsis (sepsis group) and 30 healthy individuals (control group) were enrolled,and the serum lncRNA TUG1 and HMGB1 expression levels were detected by real-time quantitative PCR,then the correlation between serum lncRNA TUG1 and HMGB1 levels was analyzed. Concentrations of inflammatory factors IFN-γ, IL-10, TNF-α and IL-6 were detected by ELISA. THP-1 macrophages were transfected into TUG1 lentivirus, which were cultured for 48 hours. Western blotting was performed to observe the expression of HMGB1 in THP-1 macrophages, MTT assay was used to detect cell viability and ELISA was used to detect concentrations of inflammatory factors. Results Compared with the control group, The serum lncRNA TUG1 expression level decreased but HMGB1 protein expression level increased in the sepsis group, and the lncRNA TUG1 level negatively correlated with HMGB1 level in the sepsis group(P<0.01). Concentrations of pro-inflammatory factors IFN-γ, TNF-α and IL-6 were significantly increased, and the level of anti-inflammatory factor IL-10 was significantly decreased. Compared with the THP-1 cells treated with LPS, the cell viability of THP-1 cells with over-expressions of TUG1 is enhanced, the expression of HMGB1 decreased, and the levels of inflammatory factors IL-6 and TNF-α were decreased. Conclusions lncRNA TUG1 inhibits the inflammatory response of sepsis, which provides a scientific basis for the diagnosis and treatment of sepsis.
