      目的:观察金钱草总黄酮对草酸钙结石模型大鼠肾组织骨桥蛋白的影响。方法:采用乙二醇饮水和氯化铵灌胃作为成石剂,30只Wistar大鼠随机分为空白对照组、草酸钙结石模型组、金钱草总黄酮组,每组10只。4周后,各组取大鼠肾脏在显微镜下观察草酸钙结晶形成情况,检测肾组织超氧化物歧化酶(SOD)、丙二醛(MDA)含量,RT-PCR检测肾组织 OPN-mRNA的表达,Western blot检测肾组织OPN蛋白的表达。结果:与草酸钙结石模型组比较,金钱草总黄酮组大鼠肾组织的结晶形成程度评分、肾小管扩张程度评分均明显降低[(2.30±0.82)比(1.60±0.70),P<0.05;3(1-4)比2(1-3),P<0.05],肾组织SOD活性显著增高[(36.64±9.77)比(52.82±12.65),P<0.01],MDA含量降低[(8.83±2.34)比(6.67±1.86),P<0.05],OPN蛋白、OPNmRNA表达水平显著降低[(0.78±0.15)比(0.47±0.13),P<0.05 ;(1.13±0.32)比(0.70±0.39),P<0.05]。结论:金钱草总黄酮具有抗氧化应激,抑制骨桥蛋白表达的作用,能有效抑制草酸钙结晶形成。
英文摘要:
      Objective: To investigate the effects of total flaconoids of Lysimachia Christinae Hance on osteopontin expression in kidney of rat model of calcium oxalate calculus. Methods: The ethylene glycol drinking water and ammonium chloride were used as stone forming agent. 30 Wistar rats were randomized to three groups, control group,calcium oxalate model group, and total flavonoids of Lysimachia Christinae Hance group, with 10 rats in each group. After 4 weeks, the formation of calcium oxalate crystallization were observed under microscope, SOD and MDA in nephridial tissues were measured, OPN mRNA was tested by RT-PCR, and OPN protein was detected by Western blot. Results: The crystallization score and renal tubular dilatation score in total flavonoids of Lysimachia Christinae Hance group were signify decreased than calcium oxalate model group [(2.30±0.82)比(1.60±0.70),P<0.05;3(1-4)比2(1-3),P<0.05], the SOD of nephridial tissue was increased [(36.64±9.77)比(52.82±12.65),P<0.01], while the MDA was reduced [(8.83±2.34)比(6.67±1.86),P<0.05], and the OPN protein and OPN mRNA expression were significantly down-regulated [(0.78±0.15)比(0.47±0.13), P<0.05; (1.13±0.32)比(0.70±0.39), P<0.05]. Conclusion: The total flavonoids of Lysimachia Christinae Hance has the effects of antioxidant stresss and inhibiting osteopontin expression, thereby it can inhibit the formation of calcium oxalate crystallization.
