      目的 探讨台州地区汉族人群5-羟色胺转运体(5-HTT)基因多态性与哮喘发生及哮喘合并抑郁之间的关联性。方法 采用聚合酶链反应技术(PCR)分别检测204例确诊哮喘患者和200例健康体检者Stin2位点的基因多态性,探讨哮喘的分子遗传学机制。采用抑郁自评量表(SDS)评测抑郁情绪,将哮喘组和健康组分为抑郁组和非抑郁组,分析哮喘组发生抑郁情况。对哮喘合并抑郁组进一步分析Stin2不同基因型及等位基因频率与抑郁发生相关性。结果 哮喘组中Stin2.10/12基因型和Stin2.10等位基因频率分别为(25.49、15.84%)与健康组具有统计学差异(P<0.05)。哮喘患者并发抑郁者88例,发生率为43.1%,健康对照组中并发抑郁发生率为17%。哮喘合并抑郁组Stin2.10/Stin2.10、Stin2.10/Stin2.12、Stin2.12/Stin2.12基因型和哮喘非抑郁组的表达具有显著性差异(P<0.01)。哮喘组Stin2基因的12、10等位基因在抑郁组的表达和非抑郁组有统计学差异(P<0.05)。结论 5-HTT基因的Stin2位点多态性可能与台州地区汉族人群哮喘发生具有相关性,是哮喘人群发生抑郁易感性的相关基因。

      Objective To investigate the association between serotonin transporter (5-HTT) gene polymorphism and asthma and asthma with depression in Han nationality in Taizhou. Methods Polymerase chain reaction (PCR) was used to detect the genetic polymorphisms of the Stin2 locus in 204 confirmed asthma patients and 200 healthy subjects, and to explore the molecular genetic mechanism of asthma. Depression was assessed by the Self-rating Depression Scale (SDS). The asthma group and healthy components were included in the depression group and the non-depression group. The depression in the asthma group was analyzed. Further analysis of the different genotypes and allele frequencies of Stin2 was associated with depression in the asthma-compressed group. Results The frequencies of Stin2.10/12 genotype and Stin2.10 allele in the asthma group were (25.49, 15.84%) and the healthy group were statistically different (P<0.05). 88 patients with asthma complicated with depression, the incidence rate was 43.1%, and the incidence of concurrent depression in the healthy control group was 17%. There were significant differences in the expression of Stin2.10/Stin2.10, Stin2.10/Stin2.12, Stin2.12/Stin2.12 genotypes and asthma non-depression group in asthma group (P<0.01). The expression of the 12 and 10 alleles of the Stin2 gene in the asthma group was significantly different between the non-depressed group and the non-depressed group (P<0.05). Conclusion The Stin2 polymorphism of 5-HTT gene may be associated with asthma in Han population in Taizhou, and it is a related gene for depression in asthmatic population.
