      摘 要 目的: 观察穴位注射结合中等强度运动对膝骨关节炎模型大鼠血清基质金属蛋白酶3(matrix metallo proteinase 3,MMP3)质量浓度的影响。方法:48只SD大鼠随机分成正常对照组、模型组、穴位注射组及注射结合跑台组。通过向SD大鼠膝关节腔内注射碘乙酸钠建立膝骨关节炎模型。正常对照组与模型组不干预,穴位注射组将鹿瓜多肽注射液注射入大鼠膝关节腔内。注射结合跑台组是在穴位注射组的基础上结合中等强度的跑台训练。分别于实验前后比较各组大鼠软骨Ⅱ型胶原含量及血清MMP3质量浓度。结果:干预6周后: ①大鼠膝关节软骨Ⅱ型胶原含量(免疫组织化学染色平均吸光度值):正常对照组(93.83±2.36),模型组(41.93±2.03),穴位注射组(54.79±2.29),注射结合跑台组(73.79±2.36)。正常对照组大鼠Ⅱ型胶原含量高于模型组(P<0.05),穴位注射组和穴位结合跑台组Ⅱ型胶原含量均高于模型组(P<0.05);穴位结合跑台组Ⅱ型胶原含量高于穴位注射组(P<0.05) ;②大鼠血清MMP3质量浓度(μg/L):正常对照组(6.69±0.17),模型组(24.68±0.56),穴位注射组(16.04±0.49),注射结合跑台组(8.89±0.39)。穴位注射组和穴位结合跑台组血清MMP3质量浓度含量均低于模型组(P<0.05);穴位结合跑台组血清MMP3质量浓度含量低于穴位注射组(P<0.05);结论:穴位注射结合中等强度运动能提高膝骨关节炎模型大鼠Ⅱ型胶原含量,并能降低模型大鼠血清MMP3质量浓度。

      ASTRACT Objective : To observe the effect of acupoint injection combined with moderate intensity exercise on the concentration of matrix metallo proteinase 3 (MMP3) in serum of knee osteoarthritis model rats. Methods: 48 SD rats were randomly divided into normal control group, model group, acupoint injection group and injection combined with running platform group. The knee osteoarthritis model was established by injecting sodium iodoacetate into the knee joint of SD rats. The normal control group and the model group did not intervene. The acupoint injection group injected deer melon polypeptide injection into the knee joint cavity of rats. Injection combined with treadmill group is based on acupoint injection group combined with moderate intensity treadmill training. Before and after the experiment, the content of type Ⅱ collagen in cartilage and the concentration of MMP3 in serum were compared. Results: after 6 weeks of intervention: ① type Ⅱ collagen content of knee joint cartilage in rats(average absorbance value of immunohistochemical staining): normal control group (93.83 ± 2.36), model group (41.93 ± 2.03), acupoint injection group (54.79 ± 2.29), injection combined with running platform group (73.79 ± 2.36). The content of type Ⅱ collagen in the normal control group was higher than that in the model group (P < 0.05), the content of type Ⅱ collagen in the acupoint injection group and the acupoint combined with the treadmill group was higher than that in the model group (P < 0.05), and the content of type Ⅱ collagen in the acupoint combined with the treadmill group was higher than that in the acupoint injection group (P < 0.05);② Mass concentration of MMP3 in serum of rats(μg/L): normal control group (6.69 ± 0.17), model group (24.68 ± 0.56), acupoint injection group (16.04 ± 0.49), injection combined with running platform group (8.89 ± 0.39). The concentration of MMP3 in the group of acupoint injection and the group of acupoint combined with running platform was lower than that in the group of model (P < 0.05), and the concentration of MMP3 in the group of acupoint combined with running platform was lower than that in the group of acupoint injection (P < 0.05); Conclusion: acupoint injection combined with moderate intensity exercise can increase the content of type Ⅱ collagen and decrease the concentration of MMP3 in serum of knee osteoarthritis rats.
