      目 的 研究微小RNA146a(miR-146a)基因rs2910164位点单核甘酸多态性与慢性阻塞性肺疾病(COPD)发生风险的相关性。方法 145例COPD患者和145例健康对照组均取5ml静脉血,采用直接测序法检测miR-146a基因rs2910164位点基因型,使用反转录荧光定量PCR(RT-PCR)检测血浆中miR-146a-5p水平,使用酶联免疫吸附法(ELISA)检测血浆IL-8细胞因子水平。结果 miR-146a基因rs2910164位点G等位基因是COPD的高风险因素(OR=1.99,95%CI=1.37-2.89)。研究组患者血浆miR-146a-5p水平显著低于对照组,血浆IL-8水平显著高于对照组,差异均有统计学意义(t=14.74,23.67;P均<0.01)。miR-146a基因rs2910164位点野生型(CC)受试者血浆miR-146a-5p水平显著高于突变型,血浆IL-8水平显著低于突变型,差异均有统计学意义(F=16.75,19.37;P均<0.01)。血浆miR-146a-5p表达水平与IL-8细胞因子水平之间呈负相关性,r=-0.63,P<0.001。结论 miR-146a基因rs2910164位点单核甘酸多态性与COPD发生风险相关,可能与miR-146a-5p负调控IL-8细胞因子表达有关。

      Objective To study the association between polymorphism of the rs2910164 locus in the microRNA146a (miR-146a) gene and the risk of chronic obstructive pulmonary disease (COPD). Methods A total of 145 COPD patients and 145 healthy controls received 5 ml of venous blood. The genotype of the rs2910164 locus of miR-146a gene was detected by direct sequencing. The miR-146a-5p levels in plasma was detected by reverse transcription-fluorescence quantitative PCR (RT-PCR). Plasma IL-8 cytokine levels were measured using enzyme-linked immunosorbent assay (ELISA). Results The G allele in miR-146a gene rs2910164 locus was a high risk factor for COPD (OR=1.99, 95% CI=1.37-2.89). The plasma miR-146a-5p level in the study group was significantly lower than that in the control group, while the plasma IL-8 level was significantly higher than that in the control group (t=14.74, 23.67; both P<0.01). The miR-146a gene rs2910164 wild-types (CC) had significantly higher plasma miR-146a-5p levels than the mutants, and plasma IL-8 levels were significantly lower than the mutants, the difference were statistically significant (F=16.75, 19.37; both P <0.01). There was a negative correlation between plasma miR-146a-5p expression level and IL-8 cytokine levels, r=-0.63, P<0.001. Conclusion The polymorphism of rs2910164 in the miR-146a gene is associated with the risk of COPD, which may be related to the negative regulation of IL-8 cytokine expression by miR-146a-5p.
