| ①目的 检测胃腺癌组织中真核翻译起始因子3a(SEukaryotic translation initiation factor 3, eIF3a)和基质金属蛋白酶-12(Smatrix metalloproteinase-12, MMP-12)的表达,分析其临床价值。②方法 选择83例胃腺癌作为观察组1,留取临床资料及术后组织,选择39例正常胃粘膜组织作为对照组1,留取术后石蜡包埋组织。选择12例胃腺癌新鲜组织作为观察组2,选择12例新鲜正常胃粘膜组织作为对照组2。应用免疫组化法检测观察组1和对照组1中eIF3a和MMP-12蛋白的表达,应用Western Bolt法检测观察组2和对照组2中eIF3a和MMP-12的半定量表达。③结果 免疫组化法显示观察组1和对照组1中eIF3a和MMP-12的表达差别有统计学意义。观察组1中eIF3a和MMP-12的表达与淋巴结转移和脉管累犯相关,eIF3a的表达与Ki67指数和肿瘤最大径相关,MMP-12的表达与间质的炎细胞浸润程度相关(P<0.05)。观察组1中eIF3a和MMP-12的表达具有正相关性,观察组1中二者的表达与生存时间相关。Western Bolt法显示观察组2和对照组2中eIF3a和MMP-12的半定量表达差别有统计学意义。④结论 胃腺癌中eIF3a和MMP-12蛋白和mRNA的表达升高,二者具有正相关性,共同促进病变的形成和进展。联合检测二者对判断预后有一定价值。 |

| ①Objective To detect expressions of Eukaryotic translation initiation factor 3(eIF3a) and matrix metalloproteinase-12(MMP-12), and investigate their relationship and clinical significance. ②Methods The study included 83 cases of gastric adenocarcinoma(clinical date and postoperative specimens) as observation group1, 39 cases of normal gastric tissue as control group1, 12 cases of fresh gastric adenocarcinoma as observation group2, 12 cases of fresh normal gastric tissue as control group2. Expressions of eIF3a and MMP-12 were detected by IHC methods in observation group1 and control group1. Expressions of eIF3a and MMP-12 mRNA were detected by Western Blot methods in observation group2 and control group2. ③Results: The positive rate of eIF3a and MMP-12 were obviously different in observation group1 and control group1 by IHC method. The positive rate of eIF3a and MMP-12 was correlated with lymph node metastasis and vascular invasion. The positive rate of eIF3a was correlated with proliferation index and maximal diameter. The positive rate of MMP-12 was correlated with inflammatory cell infiltration. There was positive relationship between eIF3a and MMP-12 in observation group1. The expressions of eIF3a and MMP-12 were correlated with prognosis in observation group1. The mRNA expressions of eIF3a and MMP-12 were obviously different in observation group2 and control group2 by Western Blot method. ④Conclusions: The higher expressions and positive correlation of eIF3a and MMP-12 may promote the occurrence and development in gastric adenocarcinoma. The joint detection of eIF3a and MMP-12 may be helpful to predict the prognosis of gastric adenocarcinoma. |
