| 目的 检测结肠腺癌中基质金属蛋白酶-11(MMP-11)、基质金属蛋白酶-12(MMP-12)和血管内皮生长因子-D(VEGF-D)的表达,分析其临床意义及相关性。方法 应用流式细胞术检测130例结肠腺癌(研究组)和130例距肿物边缘大于3厘米正常结肠粘膜组织(对照组)中MMP-11、MMP-12和VEGF-D,应用Western Blot法检测20例结肠腺癌和20例距肿物边缘大于3厘米的正常结肠粘膜组织中MMP-11、MMP-12和VEGF-D的表达。结果 结肠腺癌中MMP-11、MMP-12和VEGF-D的表达明显高于正常结肠粘膜组织,研究组中MMP-11、MMP-12和VEGF-D的表达与脉管浸润、浸润深度、淋巴结转移相关,MMP-11、MMP-12的表达与肿瘤细胞的增殖指数相关。研究组中MMP-11和VEGF-D、MMP-12和VEGF-D的表达具有正相关性。生存分析显示三者高表达患者的预后差。结论 MMP-11、MMP-12和VEGF-D在结肠腺癌中的表达上调,且具有正相关性,三者对病变的形成和进展意义明显,联合检测三者可能对判断预后有一定价值。 |

| Objective To detect the expressions of MMP-11, MMP-12 and VEGF-D in colorectal adenocarcinoma, analyze their clinical significance and correlation. Methods Protein expression of MMP-11, MMP-12 and VEGF-D were detect in 130 colorectal adenocarcinoma (study group) and 130 normal colon mucosa(control group) by FCM. Expression of MMP-11, MMP-12 and VEGF-D were detect in 20 fresh colorectal adenocarcinoma and 20 fresh normal colon mucosa by Western Blot. Results Expression of MMP-11, MMP-12 and VEGF-D was significantly different in two groups. Expressions of MMP-11, MMP-12 and VEGF-D were closely related to vascular involvement, infiltration depth and lymph node metastasis. Expression of MMP-11, MMP-12 was closely related to proliferation index. There was positive correlation between MMP-11 and VEGF-D, MMP-12 and VEGF-D. Expressions of MMP-11, MMP-12 and VEGF-D were correlated with prognosis. Conclusion Higher expressions of MMP-11, MMP-12 and VEGF-D can promote the formation and development in colorectal adenocarcinoma. MMP-11 and VEGF-D, MMP-12 and VEGF-D has a synergistic effect. Combined testing of MMP-11, MMP-12 and VEGF-D may be value in determining prognosis. |
