      目的 研究中药麻黄改善高尿酸血症大鼠模型(HUA)血尿酸(SUA)代谢作用与尿液pH的关系 方法 30只雄性SD大鼠采用随机数字表分为正常组、模型组、别嘌醇组、麻黄组及黄柏组,每组6只。采用灌胃次黄嘌呤联合腹腔注射氧嗪酸钾方法制备HUA模型,造模成功后麻黄组以麻黄水煎液20ml/(kg·d)灌胃、黄柏组以黄柏水煎液20ml/(kg·d)灌胃、别嘌醇组以混悬液20ml/(kg·d)灌胃,正常组和模型组予以等体积蒸馏水灌胃,定时定量给药7天后处死取材,测定肝脏指数(LI)、肾脏指数(KI)、SUA、血清肌酐(SCr)、24h尿量和尿pH。结果 与正常组比较:模型组和别嘌醇组KI较大[(1.10±0.07)%,(1.07±0.04)%比(0.90±0.05)%](P<0.05),麻黄组和黄柏组KI显著增加[(1.28±0.17)%,(1.18±0.14%)比(0.90±0.05)%] (P<0.01);模型组SUA水平明显升高[(77.96±19.40)比(40.66±16.77)](P<0.01)、别嘌醇组、麻黄组和黄柏组SUA无差异[(46.89±9.90)、(42.05±8.23)、(51.13±13.86)比(40.66±16.77)] (P>0.05);模型组尿量明显增加[(29.50±5.25)比(7.00±2.30)](P<0.01),别嘌醇组和黄柏组尿pH增加[(7.55±0.83),(7.63±0.73)比(6.52±0.92)] (P<0.05),麻黄组尿pH明显升高[(7.67±0.58)比(6.52±0.92)](P<0.01)。与模型组比较:麻黄组KI较高[(1.28±0.17)%比(1.10±0.072)%](P<0.05),别嘌醇组、麻黄组和黄柏组SUA水平显著降低[(46.89±9.90)、(42.05±8.23)、(51.13±13.86)比(77.96±19.40)](P<0.01),且麻黄组SUA水平与别嘌醇组和黄柏组无差异(P>0.05),麻黄组尿pH较高[(7.67±0.58)比(6.82±0.40)](P<0.05)。结论 麻黄可能通过调节尿pH发挥降低高尿酸血症模型大鼠血尿酸水平的作用。
英文摘要:
      Objective: To investigate the effect of ephedra in the treatment on hyperuricemia (HUA) rat model and elucidate the mechanism of ephedra to improve uric acid metabolism. Methods: Thirty male SD rats were randomly divided into five groups by random number table method: normal group, model group, allopurinol group, ephedra group and phellodendron amurense group, each had six individuals. HUA rat model was developed by feeding hypoxanthine and intraperitoneal injection of oteracil potassium. Rats were fed with equal volume of corresponding liquid: allopurinol group with allopurinol suspension 20ml/(kg·d), ephedra group with ephedra decoction 20ml/(kg·d), phellodendron amurense group with phellodendron amurense decoction 20ml/(kg·d), and rats in normal group and model group were fed with distilled water of the same volume. The body weight, kidney weight, liver weight, urine volume, urine pH, serum uric acid(SUA) and serum creatinine(SCr) were detected after a week of intervention. Results: Compared to the normal group: KI of model group and allopurinol group increased[(1.10±0.07)% and (1.07±0.04)% vs (0.90±0.05)%,P<0.05,repectively], KI of ephedra group and phellodendron amurense group increased significantly [(1.28±0.17)% and (1.18±0.14%) vs (0.90±0.05)%,P<0.01,respectively], level of SUA was obviously higher in model group [(77.96±19.40) vs(40.66±16.77),P<0.01], but no difference in allopurinol group, ephedra group and phellodendron amurense group[(46.89±9.90),(42.05±8.23) and (51.13±13.86) vs (40.66±16.77), P>0.05, respectively], urine volume of model increased[(29.50±5.25) vs (7.00±2.30),P<0.01] , urine pH were higher in allopurinol group and phellodendron amurense group [(7.55±0.83) and (7.63±0.73) vs (6.52±0.92) ,P<0.05, respectively] and urine pH of ephedra group was significantly increased [(7.67±0.58) vs (6.52±0.92), P<0.01]. Compared to the model group, KI of ephedra group was slightly higher [(1.28±0.17)% vs (1.10±0.072)%, P<0.05],SUA level of allopurinol group, ephedra group and phellodendron amurense group were decreased effectively [(46.89±9.90),(42.05±8.23) and (51.13±13.86) vs (77.96±19.40),P<0.01, respectively)and no difference among them (P>0.05). The urine pH of ephedra group was higher than model group’s. [(7.67±0.58) vs (6.82±0.40),P<0.05]. Conclusion: The ephedra can decrease the SUA level efficiently which is possibly contributed by alkalizing urine.
