      目的 探讨姜黄素对非酒精性脂肪性肝炎(NASH)肝细胞凋亡的影响及其相关机制。 方法 将HL-7702细胞分为对照组、模型组和治疗组,其中对照组给予普通培养基培养,模型组培养基内加入1mmol/L的游离脂肪酸(FFA)诱导NASH模型,治疗组在加入FFA的同时给予20μmol/L的姜黄素干预。48小时后收集细胞,检测细胞凋亡率、活性氧(ROS)含量及线粒体膜电位水平。 结果 模型组肝细胞的早期凋亡率及总凋亡率均明显高于对照组[(10.34±0.13)% vs.(0.55±0.08)%,P<0.05; (13.30±0.10)% vs.(2.15±0.91)%,P<0.05],治疗组与模型组相比,肝细胞早期凋亡率及总凋亡率均明显降低[(0.83±0.35)% vs.(10.34±0.13)%,P<0.05;(3.11±1.06)% vs.(13.30±0.10)%;P<0.05];模型组肝细胞内ROS含量明显高于对照组(233.58±8.00 vs. 187.24±10.60,P<0.05),而线粒体膜电位水平则明显低于对照组(1.08±0.03 vs. 1.25±0.04,P<0.05);治疗组肝细胞内ROS含量较模型组显著降低(192.57±23.86 vs. 233.58±8.00,P<0.05),而线粒体膜电位水平则显著升高(1.17±0.04 vs. 1.08±0.03,P<0.05)。 结论 姜黄素可调节NASH肝细胞凋亡,该作用可能与其线粒体保护作用相关。

      Objective To investigate the effects and mechanisms of curcumin on apoptosis of hepatocyte in nonalcoholic steatohepatitis. Methods HL-7702 cells were divided into control group, model group and treatment group. Cells in control group were cultured with standard medium while those in other two groups were incubated with 1mmol/L FFA, and 1mmol/L FFA together with 20μmol/L curcumin, respectively for 48 hours. The rate of cell apoptosis, level of ROS and mitochondrial membrane potential were measured. Results The early and total apoptosis rates in model group were significantly higher than those in control group [(10.34±0.13)% vs. (0.55±0.08)%,P<0.05; (13.30±0.10)% vs. (2.15±0.91)%,P<0.05], while the two apoptosis rates in treatment group were both alleviated [(0.83±0.35)% vs. (10.34±0.13)%,P<0.05; (3.11±1.06)% vs. (13.30±0.10)%,P<0.05]. The level of ROS was higher in model group than in control group (233.58±8.00 vs. 187.24±10.60, P<0.05), but the mitochondrial membrane potential was opposite (1.08±0.03 vs. 1.25±0.04, P<0.05). Compared with the model group, the treatment group had a lower level of ROS (192.57±23.86 vs. 233.58±8.00, P<0.05) and a higher level of mitochondrial membrane potential (1.17±0.04 vs. 1.08±0.03, P<0.05). Conclusions Curcumin can intervene apoptosis of hepatocyte in nonalcoholic steatohepatitis, which may be related to its mitochondrial protective effects.
