      目的 构建人源性DDX6多种突变体真核表达载体。方法 以DDX6-WT为模板,利用同源重组技术构分别建出2个突变体,根据突变位置,分别命名为DDX6-EQ、DDX6-△C。 结果 构建的2个突变体,经PCR、酶切、测序,2种突变完全达到预期设计,质粒构建成功。结论 成功构建2个突变体,为进一步研究DDX6在癌症进展过程中的调控作用奠定基础。
英文摘要:
      Abstract Objective To construct the different mutants of eukaryotic expression vector of human DDX6. Methods We constructed two mutants by homologous reorganization. And according to the position of the mutation, they were named as DDX6-EQ , DDX6-△C, respectively. Results The PCR, enzyme digestion, sequencing and Western Blot results showed that DDX6-EQ without helicase activity and DDX6-△C without ATPase and helicase activity were constructed. Protein molecular weights are 54kD and 34kD, respectively. Conclusion The two mutants were successfully contructed. It lay foundation for the further study of the role of DDX6 in progression of cancer.
