目的 研究槐耳清膏与顺铂联合使用对人肺癌细胞A549和A549/DDP的Bax、Bcl-2以及Survivin基因表达的影响。方法 以体外培养的A549和A549/DDP细胞株为模型,将槐耳清膏进行梯度稀释后与12μg/mL的顺铂联合使用作用于A549细胞和A549/DDP细胞,并以未使用药物刺激作为空白对照组,分别采用逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)和酶联免疫吸附法(Enzyme-Linked ImmunoSorbent Assay , ELISA)检测Bax、Bcl-2以及Survivin基因在A549细胞和A549/DDP细胞中的表达情况。结果 槐耳清膏与顺铂联合使用的情况下,随着槐耳清膏浓度的升高,A549细胞和A549/DDP细胞中Bax基因的mRNA的表达逐渐升高,Bcl-2、Survivin基因的mRNA的表达逐渐降低,差异均有统计学意义(P<0.05)。槐耳清膏与顺铂联合使用的情况下,随着槐耳清膏浓度的升高,A549细胞和A549/DDP细胞中Bax蛋白的表达量逐渐升高,Bcl-2、Survivin蛋白的表达逐渐降低,差异均有统计学意义(P<0.05)。结论 槐耳清膏与顺铂联合使用能够显著提高人肺癌细胞A549和A549/DDP的Bax基因的表达,降低Bcl-2、Survivin基因的表达。 |

Objective To study the effect of Huaier combined with cisplatin on the expression of Bax, Bcl-2 and Survivin genes in human lung cancer cell lines A549 and A549/DDP. Methods The A549 and A549/DDP cell lines were cultured in vitro. The gradient dilution Huaier and 12 μg/mL of cisplatin were added into A549 cells and A549/DDP cells. The cells untreated drugs were set as blank control. The expression of Bax, Bcl-2 and Survivin genes in human lung cancer cell lines A549 and A549/DDP were detected by RT-PCR and ELISA. Results The expression of Bax mRNA in A549 cells and A549/DDP cells increased gradually with the increase of the concentration of Huaier, and Bcl-2 and Survivin mRNA decreased with the increase of the concentration of Huaier, the difference was significant (P<0.05). The expression of Bax protein in A549 cells and A549/DDP cells increased gradually, and the expression of Bcl-2 and Survivin protein decreased gradually with the increase of the concentration of Huaier, the difference was statistically significant (P <0.05). Conclusion The combination of Huaier and cisplatin could significantly increase the expression of Bax gene, decrease the expression of Bcl-2 and Survivin genes. |
