      目 的 观察复方黄芪养心合剂(CAMNH)对冠状动脉左前降支结扎所致的大鼠缺血心肌缝隙连接蛋白CX43的影响。方法 SD大鼠50只随机分为CAMNH 大剂量组、CAMNH 小剂量组、琥珀酸美托洛尔组(MSSRT)、模型组、假手术组5组,每组10只,与相应药物灌胃后采用冠状动脉左前降支结扎术造模,结扎1h后腹主动脉取血,HE染色观察心肌细胞形态改变;采用ELISA法检测cTn-I和cTn-T的值;采用western blot(WB)法检测磷酸化的缝隙连接蛋白CX43的表达。结果 cTn-I、cTn-T:与假手术组相比模型组、MSSRT组、CAMNH小剂量组、CAMNH大剂量组均升高,且差异存在统计学意义[cTn-I:(144.94±28.83)ng/L、(85.48±24.06)ng/L、(115.07±21.34)ng/L、(81.63±16.89)ng/L比(58.50±9.53)ng/L,cTn-T:(144.33±20.21)ng/L、(97.90±11.78)ng/L、(121.33±15.02)ng/L、(100.20±5.66)ng/L比(77.40±12.52)ng/L P<0.05];MSSRT组与模型组比较升高幅度明显减少,且差异存在统计学意义(P<0.05);CAMNH大剂量组与假手术组比明显增高,与CAMNH小剂量组、模型组比增高程度减少,且差异存在统计学意义(P<0.05)。WB结果:与假手术组相比模型组、MSSRT组、CAMNH小剂量组、CAMNH大剂量组减少,且差异存在统计学意义[(0.07±0.02)、(0.13±0.01)、(0.10±0.02)、(0.13±0.02)比(0.18±0.02),P<0.05];MSSRT组与模型组比较明显增多,且差异存在统计学意义(P<0.05);CAMNH小剂量组与模型组比较略升高,差异存在统计学意义(P<0.05),与假手术组、MSSRT组相比明显减少,且差异存在统计学意义(P<0.05);CAMNH大剂量组与假手术组比明显减少,且差异存在统计学意义(P<0.05),与CAMNH小剂量组、模型组比增高程度减少,且差异存在统计学意义(P<0.05)。结论 复方黄芪养心合剂可以降低冠状动脉左前降支结扎所引起的心肌缺血的损伤,作用与琥珀酸美托洛尔缓释片相当。

      Objective To observe the effect of compound astragalus mixture nourishing heart (CAMNH) on CX43 of ischemic myocardial gap junction protein in rats caused by ligation of left anterior descending branch of coronary artery. Methods 50 SD rats were randomly divided into CAMNH high-dose group, CAMNH low-dose group, metoprolol succinate group (MSSRT), the control group, model group 5 groups, each group of 10, only with the corresponding drug lavage after by coronary artery ligation of left anterior descending branch building, 1 h after ligation of abdominal aorta in blood, HE dyeing observation myocardial cell morphology changeThe values of cTn-I and cTn-T were detected by ELISA. Western blot (WB) was used to detect the expression of phosphorylated gap junction protein CX43. Results cTn-I 、 cTn-T: Compared with the sham operation group, the model group, MSSRT group, CAMNH low-dose group and CAMNH high-dose group all increased, and the difference was statistically significant [cTn-I:(144.94±28.83)ng/L、(85.48±24.06)ng/L、(115.07±21.34)ng/L、(81.63±16.89)ng/L VS(58.50±9.53)ng/L, cTn-T:(144.33±20.21)ng/L、(97.90±11.78)ng/L、(121.33±15.02)ng/L、(100.20±5.66)ng/L VS(77.40±12.52)ng/L P<0.05]. Compared with the sham operation group, the CAMNH high-dose group was significantly higher than the CAMNH low-dose group and the model group, and the difference was statistically significant (P<0.05). WB results: Compared with the sham group, the model group, the MSSRT group, the CAMNH low-dose group and the CAMNH high-dose group were decreased, and the differences were statistically significant [(0.07±0.02)、(0.13±0.01)、(0.10±0.02)、(0.13±0.02)VS(0.18±0.02),P<0.05]. The comparison between the MSSRT group and the model group was significantly increased, and the differences were statistically significant (P<0.05). CAMNH small dose group was slightly increased compared with the model group, and the difference was statistically significant (P<0.05). Compared with the sham operation group and the MSSRT group, the difference was significantly reduced, and the difference was statistically significant (P<0.05). Compared with the sham operation group, the CAMNH high-dose group was significantly reduced, and the difference was statistically significant (P<0.05); compared with the CAMNH low-dose group and the model group, the increase was decreased, and the difference was statistically significant (P<0.05). Conclusion Compound astragalus mixture nourishing heart can reduce the injury of myocardial ischemia caused by ligation of left anterior descending branch of coronary artery, and its mechanism may be related to maintaining the phosphorylation status of myocardial gap junction protein CX43 and reducing its dephosphorylation degree, which is similar to metoprolol succinate sustained release tablet.
