      摘要 目的 观察藏红花水提液对高脂血症金黄地鼠肝脏保护的作用。方法 将48只雄性SEM金黄地鼠随机分成6组,正常对照组,模型组,藏红花水提物(低剂量组5g/kg/day 4周,中剂量组10g/kg/day 4周,高剂量组20g/kg/day 4周),阳性药组(辛伐他汀15mg/kg)。除正常对照组金黄地鼠喂食普通饲料外,其余5组均给予高脂饲料。实验结束后处死金黄地鼠,对各血脂指标、谷草转氨酶(AST)及谷丙转氨酶(ALT)水平进行检测,HE和油红O染色观察肝组织病理变化和脂质沉积情况。同时采用实时荧光定量PCR方法和蛋白质印迹法(WB)考察肝脏中PPARγ mRNA和蛋白水平。结果 与模拟组比较,藏红花水提液降低高脂血症金黄地鼠体质量[低剂量组(143.50±16.00)g、中剂量组(147.00± 7.10)g、高剂量组(138.63±14.20)g比模拟组(158.25±18.30)g],显著降低高脂血症金黄地鼠肝重[低剂量组(7.24±1.03)g、中剂量组(6.87±1.062)g、高剂量组(6.69±1.07)g比模拟组(8.51±0.96)g],显著减少总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白(LDL)水平[TC低剂量组 (18.10±3.30) mmol/L、中剂量组 (16.30±1.60) mmol/L、高剂量组(10.70±5.30) mmol/L比模拟组(20.80±4.90) mmol/L, TG低剂量组(19.40±3.00) mmol/L、中剂量组(13.50±0.90) mmol/L、高剂量组(11.90±2.30) mmol/L比模拟组(20.00±6.10) mmol/L, LDL低剂量组 (11.70±4.80) mmol/L、中剂量组(10.30±2.50) mmol/L、高剂量组(13.40±3.10) mmol/L比模拟组(17.80±3.10) mmol/L]。与模拟组比较,藏红花水提物高剂量组显著降低谷草转氨酶(AST)和谷丙转氨酶(ALT)活性[AST(108.34±29.29)比(191.79±23.53),ALT(100.71±26.92)比(144.44±37.57)]。与模型组相比,藏红花水提物各组和阳性药组中肝组织脂质沉积有所改善;藏红花水提物提高脂质代谢相关因子PPARγ mRNA和蛋白水平[低剂量组(0.64±0.13)、中剂量组(0.68±0.16)、高剂量组(1.09±0.36)比模拟组(0.40±0.05)]。结论 藏红花水提物可通过调节脂质代谢相关因子PPARγ mRNA和蛋白水平来发挥对高脂血症金黄地鼠的肝脏保护作用。

      To explore the effect of saffron on liver function in hyperlipidemic golden hamster. Methods Diet-induced hyperlipidemic golden hamsters were administered with aqueous extracts of saffron (5, 10, 20g/kg/day)and simvastatin (15mg/kg)for 4 weeks. Levels of serum triglycerides (TG), total cholesterol (TC), low-density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C) ,aspartate aminotransferase (AST) and alanine transaminase (ALT) were analyzed by biochemical analyzer. The weight of body and liver were weighed, and liver index was calculated. HE staining and Oil red O staining was used to observe the lipid accumulation in liver tissue. The proteins and mRNA expression of Peroxisome proliferator-activated receptor γ (PPARγ) in adipocyte tissue was detected by Western blot and RT-PCR. Results The results showed that saffron could decrease the levels of TC, TG, LDL-C, ALT, AST and the liver index obviously, and increase HDL-C, up-regulate the mRNA and proteins expression of PPARγ (Weight 143.50±16.00g、147.00±7.10g、138.63±14.20g vs 158.25±18.30g, Liver weight 7.24±1.03g、6.87±1.062g、6.69±1.07g vs 8.51±0.96g, TC 18.10±3.30mmol/L、16.30±1.60mmol/L、10.70±5.30mmol/L vs 20.80±4.90mmol/L, TG 19.40±3.00mmol/L、13.50±0.90mmol/L、11.90±2.30mmol/L vs 20.00±6.10mmol/L, LDL 11.70±4.80mmol/L、10.30±2.50mmol/L、13.40±3.10mmol/L vs 17.80±3.10mmol/L, AST 108.34±29.29 vs 191.79±23.53, ALT 100.71±26.92 vs 144.44±37.57, PPARγ mRNA 0.64±0.13、0.68±0.16、1.09±0.36 vs 0.40±0.05).And HE staining and oil red O staining indicated that the lipid accumulation was less in the liver tissue of the golden hamster intervented by saffron. Conclusion Aqueous extracts of saffron may exert its liver protective effect by affecting the expression of lipid metabolism related receptors PPARγ.
