      目的:探讨右归饮在治疗大鼠骨性关节炎中,诱导细胞自噬与凋亡表达的变化。方法:将SD大鼠分为右归饮治疗组、关节模型组和空白对照组,对治疗组和模型组的大鼠采用Hulth法进行膝骨关节炎造模。4周后对3组大鼠分别予右归饮、生理盐水进行灌胃治疗。在造模后第8周处死所有大鼠,取大鼠血清、膝关节及膝关节内滑膜进行Elisa、免疫印迹、HE染色及Mankin,s评分法检测,观察自噬与凋亡相关蛋白和血清中炎性因子的表达变化,以及关节软骨损伤的情况。结果:Elisa法检测见右归饮治疗组大鼠血清中炎性因子[IL-1β(20.27±1.41)pg/ml; TNF-α(162.63±10.76)pg/ml; iNOS(7.32±0.48)U/ml]的含量明显低于模型组[IL-1β(26.76±1.93)pg/ml; TNF-α(221.34±19.38)pg/ml; iNOS(10.13±0.96)U/ml],差异具有统计学意义(P <0.01),而自噬和凋亡相关蛋白的表达量在右归饮治疗组[Beclin-1(0.94±0.11); mTOR(0.16±0.09); Caspase-3(0.25±0.07); Bax(0.48±0.14)]和模型组[Beclin-1(0.54±0.08); mTOR(0.61±0.18); Caspase-3(0.60±0.12); Bax(0.79±0.11)]之间的差异也具有统计学意义(P <0.01)。HE染色及Mankin,s评分见右归饮治疗组[(5.20±0.84)分]大鼠关节软骨面较毛糙,评分低于模型组[(8.20±0.84)分],差异具有统计学意义(P <0.01)。结论:右归饮在SD大鼠膝关节滑膜细胞中可以上调自噬、下调凋亡蛋白的表达,降低血清中炎性因子的含量,缓解关节软骨的损伤。

      OBJECTIVE: This research was performed to study the effect of Yougui Drink on the treatment of osteoarthritis model rats via inducing the expression of autophagy and apoptosis. METHODS: 30 male?SD rats were randomly?divided into sham control group, osteoarthritis model group and treatment group. Rats of model group and treatment group were created into OA model, and were intragastric administrated by equivalent yougui drink and saline respectively . 8 weeks later, all the 30 rats were sacrificed, their blood was collected and the levels of inflammation factor were quantified by Elisa. Knee joints and synovial tissues were removed to be tested by western-blot and hematoxylin-eosin staining. The severity of articular cartilage damage was quantitative assessed by using the Mankin Score. RESULTS: According to data of Elisa test, levels of IL-1β(20.27±1.41 vs 26.76±1.93 pg/ml), TNF-α(162.63±10.76 vs 221.34±19.38 pg/ml) and iNOS (7.32±0.48 vs 10.13±0.96 U/ml] in treatment group decreased significantly with P <0.01 compared with model group. The expression of autophagy and apoptosis related protein[ Beclin-1(0.94±0.11); mTOR(0.16±0.09); Caspase-3(0.25±0.07); Bax(0.48±0.14) vs Beclin-1(0.54±0.08); mTOR(0.61±0.18); Caspase-3(0.60±0.12); Bax(0.79±0.11)] in treatment group are also different with model groups with P <0.01. Besides, cartilages in model group were severely damaged compared with treatment group by histopathological observation. And the result of Mankin score in model group(8.20±0.84) are higher than treatment group(5.20±0.84)(P <0.01). CONCLUSIONS: Yougui Drink treatment could attenuate articular cartilage damage via inducing autophagy upregulation and apoptosis downregulation, inhibiting inflammation factors release.
