      目的 探索元胡对小鼠乳腺癌增殖及转移的作用及机制。 方法 建立4T1乳腺癌小鼠模型,随机分为正常组、模型组、低中高剂量元胡组以及阳性药组(n=6),分别以生理盐水及不同剂量元胡提取物灌胃,阳性药组以环磷酰胺(cytoxan,CTX)腹腔注射给药,实验过程中测体重及肿瘤体积,4周后处死小鼠,检测脏器指数、瘤重、抑瘤率以及转移率;HE染色法检测胸腺、瘤体以及淋巴结形态学变化;免疫组化法检测瘤体粒细胞刺激集落因子S(granulocyte colony stimulating factor,G-CSF)、白细胞介素-10( interleukin-10,IL-10)、转化生长因子-β1(transformingSgrowthSfactor-β1,TGFβ1)和胸腺组织CD3阳性细胞的数量。 结果 结果显示元胡高剂量组肿瘤体积和重量均大于模型组,元胡高剂量组各脏器指数均高于模型组;元胡中高组肉眼可见肺转移灶数目均大于模型组;HE染色结果显示元胡各组和模型组小鼠胸腺组织的皮质、髓质区域细胞大小形态不一、排列紊乱、失去极性,细胞核畸形,呈分叶状、结节状、不规则形;染色质明显增多、增粗;而CTX组细胞排列较整齐且疏松。免疫组化结果显示,与模型组相比,元胡各组G-CSF、IL-10、TGFβ1表达增强,CD3阳性细胞数增多。 结论 元胡中高剂量组促进乳腺癌荷瘤小鼠的肿瘤增殖和转移,与元胡增加免疫器官的脏器指数,上调G-CSF、TGFβ1、IL-10等肿瘤相关炎症因子的表达和CD3阳性细胞的数量有关。因此元胡在用于晚期癌痛时,应注意控制剂量。
英文摘要:
      Objective To explore the mechanism of the effect of Yuanhu on breast cancer and its mechanism. Methods 4T1 breast cancer mouse model was established. The mice were divided into normal group, model group, Yuanhu high, Yuanhu middle, Yuanhu low and CTX group(n=6).SSAll mouthSwereStakenSbySgavage,SaStotalSdose,SblankSgroupSandSmodelSgroupSwereSgivenSdistilledSwater, CTX were peritoneally injected. The tumor weight and tumor volume were measured during the experiment. The mice were sacrificed 4 weeks later. The organ index, tumor weight, tumor inhibition rate and metastasis rate were measured. HE staining was used to detect the thymus, tumor and lymph nodes. The expression of granulocyte colony stimulating factor (G-CSF), interleukin-10 (IL-10) and transforming growth factor-β1(TGFβ1) and CD3 positive cells were measured by immunohistochemistry. Results The tumor volume and the organ index of Yuanhu high was larger than that of model group, and lung metastasis were higher than model group for Yuanhu middle and high. The results of HE staining showed that cortex and medullary region of the thymus mode and all dose of Yuanhu were different in size, disorder, loss of polarity, nucleus deformity, lobulated, nodular, irregular and chromatin significantly increased and thicker, but The cells in group CTX were arranged neatly and loosely. Immunohistochemistry results showed that all group of Yuanhu compared with the model group, G-CSF, IL-10, Yuanhu TGFβ1 and CD3 expression, the number of positive cells were increased. Conclusion The dose of middle and high of Yuanhu increased organ index of immune organs and the expression of positive cells of G-CSF, TGFβ1, IL-10 and CD3were related with Yuanhu promote tumor cell proliferation and metastasis of breast cancer. Therefore in advanced cancer, when used Yuanhu should pay attention to the dose.
