      目的:采用解毒化浊方血清干预乳腺癌SK-BR-3 细胞,观察中药血清诱导乳腺癌细胞凋亡的作用,探讨解毒化浊方治疗HER-2过表达型乳腺癌的作用机制。方法:选用雌性Wistar大鼠分为对照组、中药组、西药组、联合组四组,给药制备含药血清,给药血清分24h、48h时间干预SK-BR-3细胞,采用流式细胞技术检测中药血清干预乳腺癌细胞后细胞凋亡率。结果:药物血清干预SK-BR-3 细胞24h后,各用药组与对照组比较早期细胞凋亡率、总凋亡率明显增加,西药组、联合组尤其明显, 比较有显著性差异(P<0.05);晚期细胞凋亡率各组差异不明显,比较显著性差异(P>0.05)。干预SK-BR-3 细胞48h后,各用药组与对照组比较早期细胞凋亡率有显著性差异(P<0.05);晚期细胞凋亡率、总凋亡率比较西药组、联合组高于对照组尤其明显, 比较有显著性差异(P<0.05),西药组与联合组比较细胞凋亡率、总凋亡率比较显著性差异(P>0.05)。结论: 解毒化浊方具有诱导乳腺癌SK-BR-3细胞凋亡,协同抑制剂减毒增效的发挥抑瘤作用。

      Objective: To observe the effect of Jiedu Huazhuo decoction on human breast cancer cells SK-BR-3 and to observe the effect of traditional Chinese medicine in inducing apoptosis of breast cancer cells and to explore the effect of detoxification turbidity on HER-2 overexpression breast cancer Mechanism. Methods: Female Wistar rats were divided into 4 groups, which were respectively administrated with drug-containing serum. Serum was administered to SK-BR-3 cells for 24 h and 48 h respectively. Flow cytometry was used to detect the apoptosis rate of Chinese herbal serum after intervention of breast cancer cells. Results: Compared with the control group, the apoptosis rate and the total apoptosis rate of SK-BR-3 cells were significantly increased in the drug-treated groups compared with the control group, especially in the Western medicine group and the combined group (P < 0.05). There was no significant difference between late apoptosis rate in each group (P> 0.05). Compared with the control group, the early apoptosis rate of SK-BR-3 cells was significantly different (P < 0.05) after intervention with SK-BR-3 cells for 48h. The apoptosis rate and the total apoptosis rate of the latter groups were significantly higher than those of the western medicine group and the combined group The control group was especially obvious, with significant difference (P<0.05). Conclusion: Jiedu Huazhuo decoction can induce the apoptosis of SK-BR-3 breast cancer cells and synergize with the attenuator and synergistic effect of the inhibitor to exert antitumor effect.
