      目的观察β-catenin、GS及GPC3在肝细胞癌中的表达,探讨它们在肝细胞癌(HCC)诊断中的价值。方法 应用免疫组化EnVision法检测87例HCC、40例肝内胆管细胞癌(ICC)和70例癌旁良性肝组织中β-catenin、GS及GPC的表达。结果 ①β-catenin、GS及GPC在HCC中的阳性率分别为72.41%、87.36%和73.56%;在ICC中的阳性率分别为70.0%、40.0%和2.5%;在癌旁良性肝组织中的阳性率分别为15.71%、5.71%和2.86%;β-catenin、GS及GPC3在HCC组织中的表达显著高于癌旁良性肝组织(P<0.05);GS及GPC3在HCC组织中的表达显著高于ICC肝组织(P<0.05)。②仅GPC3在HCC不同病理分化程度有统计学意义(P<0.05)。③在HCC中β-catenin及与GPC3表达呈正相关(rs=0.229,P<0.05),β-catenin及与GS的表达不具有呈不相关性(rs=0.164,P>0.05),GS及与GPC3蛋白表达不具有相关性呈不相关(rs=0.153,P>0.05)。结论 β-catenin、GS及GPC异常表达在HCC的发生、发展中具有重要作用;GS联合GPC3有助于鉴别HCC、ICC及癌旁良性肝组织。

      Purpose To study the expression of β-catenin, GS and GPC3 in hepatocelluler carcinoma(HCC),and to explore the value of these markers in the diagnosis of HCC.Methods Immunohistochemical EnVision method was used to detect the expression of β-catenin、GS and GPC3 in HCCs, intrahepatic cholangiocarcinoma(ICC) and paracancerous benign liver tissues.Results (1)The positive rates of β-catenin、GS and GPC3 in HCCs were 72.41%, 87.36% and 73.56%,respectively,the positive rates of β-catenin、GS and GPC3 in ICCs were 70.0%,40.0% and 2.5%,but the positive rates of β-catenin、GS and GPC3 in paracancerous benign liver tissues were 15.71%,5.71% and 2.86%,respectively.The expression of β-catenin、GS and GPC3 in HCC was higher than that in benign liver tissues (P<0.05).The expression of GS and GPC3 in HCC was higher than that in ICC(P<0.05).(2)Only the positive expression of GPC3 was correlated with degree of differentiation (P<0.05). (3) The ectopic expression ofβ-catenin was significantly positively correlated with the expression of GPC3(rs=0.229,P<0.05), the ectopic expression of β-catenin was not correlated with the expression of GS(rs=0.164,P>0.05), the ectopic expression of GS was not correlated with the expression of GPC3(rs=0.153,P>0.05). Conclusion The ectopic expression ofβ-catenin, GS and GPC3 may be synergistically involved in the occurrence,and development of HCC, the combination detection of GS and GPC3 would be helpful to diagnose HCC,ICC and paracancerous benign liver tissues.
