目的 研究PTEN基因的rs2735343位点、rs112025902位点以及rs701848位点基因多态性与抑郁症患病风险之间的相关性。方法 选择2013年5月至2016年8月期间我院收治的240例抑郁症患者作为研究组,招募同期健康人240例作为对照组。取所有受试者的空腹肘静脉血5ml,抽提基因组DNA(gDNA),PCR扩增后直接测序,分析PTEN基因rs2735343位点、rs112025902位点以及rs701848位点多态性与抑郁症患病风险以及抑郁症患者抑郁症状的相关性。结果 研究组与对照组rs2735343位点、rs112025902位点以及rs701848位点各基因型和等位基因频率之间的差异均有统计学意义(P<0.05=。PTEN基因的rs2735343位点的GC+CC基因型与抑郁风险增加相关(P=0.000,OR=2.710,95%CI=1.731-4.253),该位点C等位基因与抑郁风险增加有关(P=0.006,OR=1.437,95%CI=1.097-1.884)。rs112025902位点AT+TT基因型与抑郁风险增加相关(P=0.000,OR=2.918,95%CI=1.885-4.528),该位点T等位基因与抑郁风险增加有关(P=0.000,OR=2.862,95%CI=2.183-3.754)。rs701848位点TC+CC基因型与抑郁风险增加相关(P=0.000,OR=4.498,95%CI=2.866-7.079),该位点C等位基因与抑郁风险增加有关(P=0.000,OR=3.862,95%CI=2.928-5.096)。抑郁症患者中rs2735343位点C等位基因携带者(GG+CC)的HAMD总分和与抑郁评分相关的因素的评分显著低于GG基因型携带者,差异有统计学意义(t=11.837,5.191; P=0.000,0.000)。rs112025902位点T等位基因携带者(AT+TT)的HAMD总分与焦虑因子评分显著高于AA基因型患者,rs701848位点C等位基因携带者(TC+CC)的HAMD评分与焦虑因子评分显著高于TT基因型患者,差异均有统计学意义(t=7.362,5.640; P=0.000,0.000/ t=8.010,6.051; P=0.000,0.000)。结论 PTEN基因的rs2735343位点、rs112025902位点以及rs701848位点多态性可能与抑郁症以及抑郁症状的风险相关。 |

Objective To study the correlation between PTEN gene polymorphism of rs2735343, rs112025902 and rs701848 locus and the risk of depression. Methods A total of 240 patients with depression from May 2013 to August 2016 were selected as the study group, and another 240-healthy people were recruited as the control group. 5 ml of fasting elbow vein blood of all subjects were taken and genomic DNA (gDNA) extracted. After PCR amplification and direct sequencing, the correlation between PTEN gene polymorphism of rs2735343, rs112025902 and rs701848 locus and the risk of depression were analyzed. Results There was significant difference between rs2735343 locus, rs112025902 locus and rs701848 locus genotype and allele frequency in the study group and control group (P <0.05). The GC + CC genotype of the rs2735343 locus of the PTEN gene was associated with an increased risk of depression (P = 0.000, OR = 2.710, 95% CI = 1.731-4.253), and the C allele of the rs2735343 locus was associated with an increased risk of depression (P = 0.006, OR = 1.437, 95% CI = 1.097-1.884). AT + TT genotype of rs112025902 locus was associated with an increased risk of depression (P = 0.000, OR = 2.918, 95% CI = 1.885-4.528), and the T allele of the rs112025902 locus was associated with an increased risk of depression (P = 0.000, OR = 2.862, 95% CI = 2.183-3.754). The TC+CC genotype of rs701848 locus was associated with an increased risk of depression (P = 0.000, OR = 4.498, 95% CI = 2.866-7.079), and the C allele of rs701848 locus was associated with an increased risk of depression (P = 0.000, OR = 3.862, 95% CI = 2.928-5.096). The scores of the HAMD score and factors associated with depression scores of the rs2735343 locus C allele (GG + CC) carriers in the depressive patients were significantly lower than in the GG genotype carriers (t = 11.837, 5.191; P = 0.000, 0.000). The HAMD total score and anxiety factor score of rs112025902 locus T allele (AT + TT) carriers were significantly higher than AA genotype carriers, the HAMD total score and anxiety factor scores of rs701848 locus C allele carriers (TC + ??CC) were significantly higher than TT genotype carriers, the differences were statistically significant (t = 7.362, 5.640; P = 0.000, 0.000/t = 8.010, 6.051; P = 0.000, 0.000). Conclusion The rs2735343, rs112025902 and rs701848 polymorphisms of the PTEN gene may be associated with the risk of depression and depressive symptoms. |
