      【】目的 通过观察NOD1和NOD2在哮喘大鼠肺组织中的表达,探讨冬虫夏草治疗哮喘的可能作用机制。方法 通过哮喘大鼠模型,给予冬虫夏草灌胃,分别采用免疫组化法和实时荧光定量PCR法检测肺组织NOD1和NOD2蛋白或mRNA的表达。 结果 哮喘组(0.62±0.34)NOD1 mRNA的表达量显著低于对照组(1)(P﹤0.01);冬虫夏草组(0.98±0.22)NOD1 mRNA的表达量显著高于哮喘组(0.62±0.34)(P﹤0.01)。哮喘组NOD2蛋白(0.18±0.03 OD值)的光密度值显著低于对照组(0.24±0.03 OD值)(P﹤0.01),而哮喘组(0.92±0.32)NOD2 mRNA的表达量与对照组(1)差异无统计学意义(P>0.05);冬虫夏草组(依次为:0.23±0.02 OD值;1.27±0.48)NOD2蛋白和 mRNA的表达量显著高于哮喘组(依次为:0.18±0.03 OD值;0.92±0.32)(均P﹤0.05)。NOD2 mRNA和蛋白的表达水平无显著相关性 ( n=24, r =0.32,P>0.05);NOD1mRNA和NOD2 mRNA的表达水平无显著相关性( n=24, r =0.20,P>0.05)。结论 哮喘大鼠肺组织 NOD1和NOD2的表达下降,冬虫夏草能上调NOD1和NOD2表达的作用。

      Objective To investigate the potential roles of cordyceps sinensis on asthma therapeutics pathogenesis, the nucleotide-binding oligomerization domain-like receptor (NOD)1 and 2 was observed. Methods Rat asthma model was challenged, cordyceps sinensis was given by intragastric administration. The expressions levels of NOD 1 and 2 protein or mRNA were detected by immunohistochemisty or real-time PCR methods at lung tissue. Results Levels of NOD1 mRNA in asthma group (0.62±0.34 ) was significantly lower than that in control group(1) (P﹤0.01).Dramaticly, levels of NOD1 mRNA in cordyceps sinensis group(0.98±0.22) was significantly increased than that in asthma group (0.62±0.34 )(P﹤0.01). Moreover, levels of NOD2 protein in asthma group (0.18±0.03 optical density) was significantly lower than that in control group(0.24±0.03 optical density)(P﹤0.01), while there was no statistical significance of NOD2 mRNA in asthma group(0.92±0.32)when compared to control group(1)(P>0.05). Farthermore, levels of NOD2 protein and mRNA in cordyceps sinensis group (0.23±0.02 optical density, 1.27±0.48, respectively)were significantly higher than those in asthma group(0.18±0.03 optical density;0.92±0.32)(both P<0.05). And there was no significance correlation neither between NOD2 protein and mRNA ( n= 48, r =0.32,P>0.05) nor between NOD1 mRNA and NOD2 mRNA( n=24, r =0.20,P>0.05). Conclusion Levels of NOD1 and 2 was decreased in asthmatic rats lung tisue, while it can be up-regulated by cordyceps sinensis.
