      目的:探讨疏肝健脾活血方含药血清对活化的大鼠肝星状细胞TGF-β1/Smads信号通路相关基因表达的影响。方法:沉默Smad4基因得到Smad4 RNAi HSCs-T6细胞株后与HSCs-T6细胞株分别随机分组,HSCs-T6细胞株分为空白对照组(N组)、TGF-β1模型组(T组)、TGF-β1 含药血清组(TH组);Smad4 RNAi HSCs-T6细胞株分为Smad4 RNAi HSCs-T6细胞分组为:Smad4 RNAi组(S组)、 Smad4RNAi TGF-β1模型组(ST组)、Smad4 RNAi TGF-β1 含药血清组(STH组),用含药血清干预后采用ELISA法检测大鼠肝星状细胞中Ⅰ型胶原含量的变化;RT-PCR法检测HSC-T6细胞中Smad2、Smad3、Smad4、Smad7 mRNA的相对表达量。结果:TGF-β1促进HSC的I型胶原及Smad2、Smad3、Smad7的表达(P<0.01或P<0.05),含药血清干预下HSC的I型胶原及Smad2、Smad3、Smad4、Smad7表达下调(P<0.01或P<0.05),沉默Smads4基因后得到相同结果(P<0.01或P<0.05)。结论:疏肝健脾活血方在离体条件下可以延缓肝纤维化的进程,其机制可能与阻断TGF-β1/Smads信号通路有关。
英文摘要:
      Objective:To explore the effect of serum containing Shugan Jianpi Huoxue Decoction on the expression of TGF-β1/Smads pathway genes in activated rat HSC. Methods: Smad4 RNAi HSCs-T6 were obtained by using lentiviral vector to silent Smad4 gene in HSCs-T6. Then HSCs-T6 and Smad4 RNAi HSCs-T6 were randomly grouped, respectively.The specific groups were as follows:HSCs-T6 were divided into control group(N),model group(T) and serum containing medicine group(TH);Smad4 RNAi HSCs-T6 were divided into Smad4 RNAi group(S), Smad4 RNAi model group(ST) and Smad4 RNAi serum containing medicine group(STH). After the intervention of serum containing medicine, the collagenⅠ in HSCs was measured by ELISA ;RT-PCR was used to detect the relative expression of Smad2, Smad3, Smad4 and Smad7 mRNA in HSC. Results:TGF-β1 significantly promotes the expression of collagenⅠ and genes of Smad2/Smad3/Smad7 in HSC(P<0.01 or P<0.05).After the intervention of serum containing medicine,the expression of collagenⅠ and genes of Smad2/Smad3/Smad4/Smad7 were significantly declined(P<0.01 or P<0.05), as well as the silence of Smad4 gene(P<0.01 or P<0.05). Conclusion:Shugan Jianpi Huoxue Decoction can retard the liver fibrosis in vitro, and the mechanism may be related to blocking TGF-β1/Smads pathway.
