| 目的 研究血管内皮生长因子（vascular endothelial growth factor，VEGF）基因多态性与负性生活事件之间的相互作用对重度抑郁症易感性的影响。方法 选择85例我院收治的重度抑郁症患者作为研究组，根据研究组患者的年龄、性别匹配招募85例健康人作为对照组，抽提所有受试者的外周血2ml后提取基因组DNA，采用直接测序法对VEGF基因rs2010963位点的多态性进行分析。采用生活事件量表（the Life Events Scale，LES）评估负性生活时间的频率和严重程度，并采用Logistic回归方法评估基因与环境的相互作用。结果 研究组与对照组VEGF基因rs2010963位点各基因型频率之间的差异具有统计学意义（χ2=7.201，P=0.027），但是各等位基因频率之间的差异没有统计学意义（χ2 =0.963，P=0.327）。VEGF基因rs2010963位点C等位基因携带者与较高的负性生活事件之间的相互作用显著，P=0.002，OR=4.832, 95%CI=1.522~15.877。结论 VEGF基因rs2010963多态性与环境之间的相互作用增加了重度抑郁症发生的风险，携带C等位基因的个体在经历高负性生活事件时更容易发生重度抑郁症。 |

| Objective To study the effect of the interaction between vascular endothelial growth factor (VEGF) gene polymorphism and negative life events in the susceptibility to major depressive disorder(MDD). Methods A total of 85 MDD patients were selected as the study group, and 85 healthy subjects were recruited as the control group according to the age and gender matching of the study group. The genomic DNA was extracted from the peripheral blood of all subjects. The polymorphism of rs2010963 locus in VEGF gene was analyzed by direct sequencing. The frequency and severity of negative life events were assessed by the Life Events Scale (LES), and the interaction of genes with the environment was assessed by Logistic regression. Results There was a significant difference in the genotype frequencies of rs2010963 locus between the study group and the control group (χ2 = 7.201, P = 0.027), but there was no significant difference between the allele frequencies (χ2 = 0.963, P = 0.327). The interaction between rs2010963 locus C allele carriers and higher negative life events was significant, P = 0.002, OR = 4.832, 95% CI = 1.522 ~ 15.877. Conclusion The interaction between the rs2010963 polymorphism of VEGF gene and the environment increases the risk of major depressive disorder. Individuals carrying the C allele are susceptible to major depressive disorder when suffering from high negative life events. |
