      摘要:目的 观察黄葵胶囊对慢性肾小球肾炎大鼠炎症因子和免疫功能的影响。方法 采用 小牛血清白蛋白法建立慢性肾小球肾炎大鼠模型,将40只大鼠模型随机分为模型组、黄葵胶囊低、中、高剂量组,每组10只,另设10只大鼠作为对照组,对照组和模型组给予生理盐水灌胃治疗,黄葵胶囊低、中、高剂量组分别给予0.5g·kg-1、1.0g·kg-1和2.0g·kg-1的黄葵胶囊灌胃。8周后比较各组大鼠的肾组织病理学变化、血清肌酐(Scr)、尿素氮(BUN)、尿酸(UA)含量,白介素-6(IL-6)、细胞间粘附因子-1(ICAM-1)、转化生长因子-β1(TGF-β1)水平和外周血T淋巴细胞亚群比例。结果 对照组肾小球、系膜基质未出现明显病理变化,模型组肾小球硬化、系膜基质增生,黄葵胶囊各剂量组肾脏病理变化较模型组明显减轻。模型组大鼠血清Scr、BUN和UA含量高于对照组,但明显低于黄葵胶囊各剂量组,随黄葵胶囊剂量的增加含量降低,差异均有统计学意义(P<0.05)。模型组大鼠IL-6、ICAM-1和TGF-β1水平均高于对照组,且明显高于黄葵胶囊低、中、高剂量组,随黄葵胶囊剂量的增加水平降低,差异均有统计学意义(P<0.05)。模型组大鼠外周血CD3、CD4和CD4/CD8均明显低于对照组和黄葵胶囊组,CD8高于对照组和黄葵胶囊组,随黄葵胶囊剂量的增加外周血T淋巴细胞亚群比例趋于稳定,差异均有统计学意义(P<0.05)。结论 黄葵胶囊可保护慢性肾小球肾炎大鼠肾脏组织,改善肾功能指标,减轻炎症反应,机制可能与调节免疫相关。

      Abstract: Objective To observe the influence of Huangkui capsule on inflammatory factor and immune function in chronic glomerulonephritis rats. ambrette capsule on chronic glomerulonephritis inflammatory factor and the influence of immune function in rats. Methods The chronic glomerulonephritis rats model were established by calf serum albumin method, l 40 rats mode were randomly divided into model group, huangkui capsule low, medium and high dose group, each group with 10 rats, and other 10 rats were set as control group. The control group and model group were given normal saline, and the experimental groups were given 0.5g·kg-1, 1.0g·kg-1 and 2.0g·kg-1 huangkui capsule. After 8 weeks, the renal histopathological changes, content of serum creatinine (Scr), blood urea nitrogen (BUN) and uric acid (UA), level of interleukin-6 (IL-6), intercellular adhesion factor-1 (ICAM-1) and transforming growth factor-beta 1 (TGF-β1), and the ratio of peripheral blood T lymphocyte subsets were compared. Results There were not appear obvious pathological changes of glomerular, mesangial matrix in the control group, , mesangial matrix were proliferation, glomerular sclerosis in the model group, the kidney pathological changes in the experimental group were significantly reduce groups compared with model group. The contents of Scr, BUN and serum UA in the model group were higher than the control group, were significantly lower than the experimental group, with the increase of huangkui capsule dose, the contents were lower, the differences were statistical significance (P < 0.05). The level of IL-6, ICAM-1 and TGF-β1 in the model group were higher than the control group, were significantly higher than the experimental group, with the increase of huangkui capsule dose, the levels were reduced, the differences were statistically significant (P < 0.05). The ratios of peripheral blood T lymphocyte subsets CD3, CD4 and CD4/CD8 in the model group were lower than the control group and experimental group, the ratio of CD8 was higher than the control group and experimental group, with the increase of huangkui capsule dose, the ratios of peripheral blood T lymphocyte subsets were tended to be stable, the differences were statistical significance (P < 0.05). Conclusion Huangkui capsule can protect the kidney tissue of chronic glomerulonephritis rats, improve renal function indexes, reduce inflammation, and the mechanism may be associated with immune regulation.
