      目的 明确山茱萸新苷是否可以抑制实验性自身免疫性脑脊髓炎(EAE)大鼠中枢神经系统中免疫细胞浸润及血管细胞粘附分子-1(VCAM-1)的表达。 方法 制作EAE模型。32只Lewis大鼠被随机均分为正常对照组、模型组、强的松组以及山茱萸新苷组,每天评定神经功能缺损症状,当症状达到高峰期时处死所有大鼠,采用HE染色法比较每组大鼠免疫细胞浸润中枢神经系统的严重程度,实时荧光定量PCR(RT-PCR) 比较VCAM-1 mRNA的表达,Western Blotting比较VCAM-1蛋白的表达。 结果 上述4组大鼠第14d神经功能缺损症状评分分别为0分、(3.83±0.98)分、(0.67±0.82)分、(1.67±1.75)分,HE染色病灶个数分别为0、8.75±2.23、2.33±1.03、4.83±1.47,VCAM-1 mRNA的相对表达量分别为5.602±0.761、37.762±4.560、17.143±1.521、28.042±3.147,VCAM-1蛋白的相对表达量分别为7.958±2.265、22.433±1.829、15.725±1.338、18.625±0.874。与模型组大鼠相比较,山茱萸新苷组大鼠有较低的神经功能缺损症状评分,较少的中枢神经系统病灶个数,较低的VCAM-1 mRNA相对表达量及VCAM-1蛋白的相对表达量(P=0.002、0.037、0.012、0.004)。 结论 山茱萸新苷可以缓解EAE大鼠的神经功能缺损症状,可以抑制免疫细胞浸润EAE大鼠中枢神经系统,可以抑制EAE大鼠中枢神经系统中VCAM-1 mRNA及蛋白质的表达。

      Objective To determine whether cornuside can suppress invasion of immune cells and expression of VCAM-1 in the central nervous system in rats which suffered from experimental autoimmune encephalomyelitis (EAE). Methods EAE model was prepared. 32 Lewis rats were divided into normal control group, model group, prednisone group and cornuside group randomly. We evaluated the neurological deficit symptoms of rats every day, and all rats were sacrificed when the neurologic deficits reached peak stage. We adopted HE staining to compare the severity of the invasion of immune cells, real time quantitative PCR (RT-PCR) to compare the expression of VCAM-1 mRNA, and Western Blotting to compare the expression of VCAM-1 protein in each group. Results The scores of neurological deficit symptom of Lewis rats in the four groups mentioned above were respectively 0, 3.83±0.98, 0.67±0.82, 1.67±1.75 on fourteenth day, the number of lesions stained by HE were respectively 0, 8.75±2.23, 2.33±1.03, 4.83±1.47. The relative expression of VCAM-1 mRNA were respectively 5.602±0.761, 37.762±4.560, 17.143±1.521, 28.042±3.147, and the relative expression of VCAM-1 protein were respectively 7.958±2.265, 22.433±1.829,15.725±1.338, 18.625±0874. Compared with model group rats cornuside group rats had lower score of neurological deficit symptoms, less number of lesions, lower relative expression of VCAM-1 mRNA and protein in the central nervous system (P=0.002, 0.037, 0.012, 0.004). Conclusion Cornuside can alleviate neurological deficit symptoms, suppress invasion of immune cells, and inhibit the expression of VCAM-1 mRNA and protein in the central nervous system in rats with EAE.
