      目的: 探讨中药活性成分桔梗皂苷D是否对阿霉素有协同抗肝癌效应并研究其机制。方法: MTT实验检测肝癌细胞系PLC的细胞活力;流式细胞术检测PLC细胞的凋亡和活性氧簇(ROS)的产生;western blot实验检测PLC细胞中ASK1及JNK的磷酸化水平及caspase-3的活化水平。结果: 阿霉素联合桔梗皂苷D对PLC的细胞活力抑制率(63.2±5.4)和凋亡诱导率(35.1±3.1)显著高于阿霉素单治疗组的的细胞活力抑制率(16.6±1.3,P<0.05)和凋亡诱导率(8.2±0.7,P<0.05)。桔梗皂苷D处理不影响PLC细胞内的ROS水平,但阿霉素联合桔梗皂苷D处理组的ASK1及JNK的磷酸化水平及caspase-3的活化水平均显著高于阿霉素单治疗组。另外,阿霉素+桔梗皂苷D+ASK1小干扰RNA组PLC的细胞活力抑制率(21.5±1.8)和凋亡诱导率(11.3±0.9)显著低于阿霉素联合桔梗皂苷D组PLC的细胞活力抑制率(63.2±5.4,P<0.05)和凋亡诱导率(35.1±3.1,P<0.05)。结论: 桔梗皂苷D上调ASK1的表达增强阿霉素的体外抗肝癌活性。

      AIM: To investigate the synergistic effect and mechanism of platycodin D on increasing the cytotoxicity of adriamycin to hepatocellular carcinoma. Methods: MTT assay was performed to measure the cell viability of PLC. Flow cytometry analysis was performed to detect the apoptosis and production of ROS in PLC cells. Western blot analysis was performed to evaluate ASK1 and JNK phosphorylation, and the activation of caspase-3 in PLC cells. Results: Cell viability inhibitory rate (63.2±5.4) and apoptotic rate (35.1±3.1) of PLC co-treated with adriamycin and platycodin D was significantly higher than the cell viability inhibitory rate (16.6±1.3, P<0.05) and apoptotic rate (8.2±0.7, P<0.05) of PLC cells treated with single adriamycin. platycodin D treatment can not change the production of ROS in PLC. However, the phosphorylation of ASK1 and JNK, and the activation of caspase-3 was significantly higher in the adriamycin plus platycodin D group rather than the adriamycin single treatment group. In addition, cell viability inhibitory rate (21.5±1.8) and apoptotic rate (11.3±0.9) of PLC in adriamycin + platycodin D + ASK1 small interfere RNA group was significantly lower than the cell viability inhibitory rate (63.2±5.4, P<0.05) and apoptotic rate (35.1±3.1, P<0.05) of PLC in adriamycin + platycodin D group. Conclusion: Platycodin D enhances the anti-tumor effect of adriamycin on hepatocellular carcinoma in vitro by upregulating the expression of ASK1.
