目的:观察五福饮颗粒剂(WFYG)对膝骨关节炎大鼠膝关节的影响,为从“五脏同补”角度治疗膝骨关节炎提供依据。方法:将SPF级雄性大鼠30只随机分为假手术组、模型组和治疗组,每组各10只;模型组和治疗组均采用Hulth法建立大鼠KOA模型;造模6周后治疗组大鼠开始灌胃WFYG,假手术组和模型组灌胃等量生理盐水,共4周。采用ELISA测定WFYG对KOA大鼠血清及关节液中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的影响,采用IHC法观察WFYG对KOA大鼠软骨中骨形态发生蛋白-2(BMP-2)的影响。结果:与模型组相比,WFYG可以显著降低血清IL-1β[(2.751±0.269)ug/l比(3.660±0.371)ug/l,P<0.01]、IL-6[(2.719±0.391)ug/l比(3.385±0.313)ug/l,P<0.01]含量,以及关节液中IL-1β[(2.670±0.381)ug/l比(3.630±0.384)ug/l,P<0.01]、IL-6[(2.517±0.343)ug/l比(3.254±0.460)ug/l,P<0.01]含量,上调BMP-2[(0.371±0.025)比(0.212±0.018),P<0.01]的表达。结论:五福饮颗粒剂可通过降低KOA大鼠血清和关节液中IL-1β、IL-6的含量,上调关节软骨中BMP-2的表达,延缓关节软骨退变。 |

Objective To observe the influence of Wufuyin Granule at knee-joints of rats with knee osteoarthritis,and provide the basis for “Tonifying Five Zang-organs”to treat knee osteoarthritis.Methods Thirty male SPF(Specific Pathogen Free)rats were randomly divided into the sham operation control group(SC group),the ischemia model group(IC group)and the Wufuyin Granule preventive group(WFYGP group),and each group had ten rats. The IC group and the WFYGP group received modified Hulth's operation to induce KOA model,and then the WFYGP group was treated with WFYG 6 weeks later after mading model,once per day,and the SC group and the IC group were managed with same amount of normal saline.After 4 weeks,ELISA was used to detect the effect of WFYG on IL-1β、IL-6 in the serum and synovial fluid. Immunohistochemisty was used to detect the effect of WFYG on the BMP-2 in articular cartilage. Results Compared with IC group,WFYG could significantly reduce serum IL-1[(2.751±0.269)ug/l VS(3.660±0.371)ug/l,P<0.01]、IL-6[(2.719±0.391)ug/l VS(3.385±0.313)ug/l,P<0.01] content,and synovial fluid IL-1[(2.670±0.381)ug/l VS(3.630±0.384)ug/l,P<0.01]、IL-6[(2.517±0.343)ug/l VS(3.254±0.460)ug/l,P<0.01] content,up regulate the expression of BMP-2 [(0.371±0.025)VS(0.212±0.018),P<0.01]. Conclusion Wufuyin Granule can reduce the content of IL-1β、IL-6 in serum and synovial fluid,up regulation of BMP-2 expression in articular cartilage,delaying the degeneration of articular cartilage. |
