PI3K作为一种胞内磷脂酰肌醇激酶,是细胞内重要的信号转导分子,参与调节细胞的增殖、凋亡与分化等生理过程。众多研究表明PI3K依赖性信号通路(以PI3K-Akt-mTOR通路为主)的过度激活与肿瘤的发生有密切关系。过去的几十年中以PI3K-Akt-mTOR通路为靶点的治疗已经取得了很大进步,使得人们认识到PI3K抑制剂在肿瘤的治疗中发挥的重要作用。本文将对PI3K抑制剂在抗肿瘤方面的研究进展作一综述。 |

PI3K, an intracellular phosphatidylinositol kinase, plays an important role in the regulation of cell proliferation, apoptosis and differentiation. Numerous studies have shown that the excessive activation of PI3K-dependent signaling pathway (mainly PI3K-Akt-mTOR pathway) is closely related to tumorigenesis. The treatment targeting PI3K-Akt-mTOR pathway has made great progress in the past several decades, and it has been recognized that PI3K inhibitors play an important role in tumor therapy. In this paper, the progress of PI3K inhibitors in anti-tumor research is reviewed. |
