      目的 研究血管内皮生长因子对骨髓间充质干细胞在大鼠离体心脏中滞留率的影响。方法 取100只雌性大鼠随机分成正常灌注组、正常灌注+血管内皮生长因子组、缺血再灌注组、缺血再灌注+血管内皮生长因子4组各25例开展实验。用荧光定量聚合酶链反应(RT-PCR)检测后得出各个不同时间点的骨髓间充质干细胞滞留率。结果 发现骨髓间充质干细胞滞留率分别在缺血再灌注组(加血管内皮生长因子组)著性大于正常灌注组(加血管内皮生长因子组);经血管内皮生长因子预处理和没有预处理对其滞留率无显著性差异。结论 缺血再灌注可以显著增加大鼠离体心脏的骨髓间充质干细胞滞留率;而血管内皮生长因子对其无明显增加作用。
英文摘要:
      Objective To study the retention rate of bone marrow mesenchymal stem cells (BMSCs) in normal vitro perfusion model and ischemical reperfusion injury model after the infusion of BMSCs with/without vascular endothelial growth factor(VEGF) in rats. Methods 100 female rats were divided into 4 groups: normal vitro perfusion group( A1), normal vitro perfusion +VEGF group(A2), ischemical reperfusion injury group( B1), ischemical reperfusion injury+VEGF group(B2). All were infused with purified BMSCs 1ml (1.0x106/ml) of male rats ; there was infused with VEGF 1ml (1000μmol/L) before use of BMSCs In group A2 and B2. The retention rate of BMSCs was calculated for counting the number of SRY in Y chromosome by RT-PCR in each group within 5、15、30、60、120min. Results The retention rate of BMSCs was 16.32±3.839%、14.02±5.231%、12.48±3.456%、12.42±3.608%、12.42±3.491% in groupA1; 19.38±4.660%、19.54±5.108%、16.68±2.639%、16.06±2.124%、15.60±2.003% in group A2; 36.96±4.795%、35.88±4.762%、28.05±3.459%、27.04±6.352%、27.06±5.838% in group B1; 41.46±5.878%、37.82±5.737%、32.82±3.534%、33.24±6.191%、31.60±3.259% in group B2 at every time points ; so it was significant higher in group B1 compared with group A1 the same as group B2 compared with groupA2, but it wasn’t significant different between group A1and A2 like as group B1 and B2. Conclusion The retention rate of BMSCs was improved much more in ischemical reperfusion injury model than normal vitro perfusion model. but it didn’t increased obviously after the infusion of BMSCs with the preconditioning of VEGF as vascular permeability.
