      摘要:目的:观察姜黄素对溃疡性结肠炎(Ulcerative Colitis,UC)模型小鼠结肠组织肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素10(IL-10)表达的影响,探讨其治疗UC的可能作用机制。方法:将60只鼠龄8周,体质量(20±2)g,SPF级健康雄性BALB/c小鼠随机分为正常对照组、模型对照组、姜黄素治疗组、柳氮磺胺吡啶(salazosulfapyridine,SASP)治疗组4组。采用右旋葡聚糖硫酸钠(dextran sulphate sodium,DSS)法复制UC小鼠模型后,每日腹腔注射给药1次,连续7天。实验期间每日观察小鼠一般生理状态并记录,行疾病活动度指数(disease activity index, DAI)评分,末次给药后处死小鼠,解剖观察并进行结肠大体形态损伤指数(colon macroscopic damage index, CMDI)评分、组织切片染色,光镜下观察结肠组织的病理学变化并评估组织损伤指数(tissue damage index, TDI)。ELISA法检测小鼠结肠组织中促炎因子TNF-α、IL-1β和抗炎因子IL-10的表达水平。结果:①模型组大体形态、组织病理均符合溃疡性结肠炎特点。②各组DAI评分比较:空白组(0.06±0.05)分,模型组(1.38±0.61)分, SASP组(0.51±0.40)分,姜黄素组(0.52±0.36)分,SASP组、姜黄素组与模型组比较,P<0.01。③各组CMDI评分比较:空白组(0.69±0.38)分,模型组(3.83±1.44)分,SASP组(2.14±0.92)分,姜黄素组(2.19±1.10)分,SASP组、姜黄素组与模型组比较,P<0.01。④各组TDI评分比较:空白组(4.14±1.12)分,模型组(8.89±0.88)分, SASP组(5.48±1.01)分,姜黄素组(6.48±1.03),SASP组、姜黄素组与模型组比较,P<0.01。⑤各组小鼠结肠组织的TNF-α、IL-1β及IL-10含量比较:空白组(28.99±12.95、43.69±11.88、166.58±13.26),模型组(418.19±41.67、401.11±35.32、33.96±6.68), SASP组(126.75±6.16、157.51±20.43、156.33±16.51),姜黄素组(254.83±28.29、236.83±25.21、136.66±16.49),SASP组、姜黄素组与模型组比较,P<0.01。结论:姜黄素可以显著改善UC小鼠症状、病理形态,上调IL-10表达水平,抑制TNF-α、IL-1β表达水平,达到对UC的抗炎作用。

      Abstract: Objective: To observe the effects of curcumin on the expression of TNF-α、IL-1βand IL-10 of colon tissue in mice with Ulcerative Colitis(UC), and to explore the possible mechanisms of its therapeutic effects on UC. Methods: The 60 rats were aged 8 weeks, and the body weight was (20±2) g, SPF grade healthy male BALB/c mice were randomly divided into four groups: blank control group, model control group,curcumintreatmentgroup,salazosulfapyridine(SASP)/positive control group. UC model was established by dextran sulphate sodium(DSS), and then daily intraperitoneal injection administration was given for 7 days. During the course of experiment, the general conditions of mice were observed, and disease activity index (DAI) were assessed. At the end of treatment, colon tissue were dissected, its macroscopic damage index (CMDI) and tissue damage index(TDI) were evaluated, TNF-α, IL-1β and IL-10 were determined by ELISA. Results:① General morphology and histopathology of the model group conforms to the manifestations of ulcerative colitis. ② Comparison of DAI score: blank group (0.06±0.05), model group (1.38 ± 0.61), SASP group (0.51 ± 0.40), curcumin group (0.52 ± 0.36), significant difference was found between SASP group and curcumin group compared with model group, P<0.01. ③Comparison of CMDI score : blank group (0.69 ± 0.38), the model group (3.83 ± 1.44), SASP group (2.14± 0.92), curcumin group (2.19±1.10), significant difference was found between SASP group and curcumin group compared with model group, P<0.01. ④Comparison of TDI score : blank group (4.14 ±1.12), the model group (8.89 ± 0.88), SASP group (5.48 ±1.01), curcumin group (6.48 ±1.03) , significant difference was found between SASP group and curcumin group compared with model group, P<0.01.⑤ Comparison of TNF-α,IL-1βand IL-10 contents in colon tissue of mice: the blank group (28.99 ±12.95, 43.69 ± 11.88, 166.58±13.26), model group (418.19±41.67, 401.11±35.32, 33.96 ±6.68), SASP group (126.75±6.16, 157.51±20.43, 156.33 ±16.51), curcumin group (254.83±28.29, 236.83 ± 25.21, 136.66±16.49) , significant difference was found between SASP group and curcumin group compared with model group, P<0.01. Conclusion: curcumin can significantly improve the symptoms and pathological morphology of UC mice, by regulating the expression of IL-10 and inhibiting the expression of TNF-α、IL-1β, through which achieving the anti-inflammatory effect of UC.
