目的:研究葛根素对氧诱导大鼠视网膜新生血管VEGF和PECAM-1表达影响,进一步探讨其对新生血管的抑制作用。
方法:建立氧诱导大鼠新生血管模型,并分成空白组,葛根素小剂量组及葛根素大剂量组(每组4只),通过荧光素眼底血管造影观察各组新生血管渗漏区域改变,免疫荧光染色观察PECAM-1表达,ELISA测定玻璃体VEGF表达量。
结果:新生血管渗漏程度、PECAM-1,VEGF表达随葛根素剂量增加而减少,与空白组比较,葛根素小剂量玻璃体液中 VEGF 表达明显降低( p<0.001), 且葛根素大剂量组降低程度明显增大(p<0.001)。
结论:葛根素可通过抑制VEGF及PECAM-1表达,抑制氧诱导大鼠视网膜新生血管增生。 |

ABSTRACT: Objective: To investigate the effect of puerarin on the expression of VEGF and PECAM-1 on hyperoxia-induced retinopathy in rats. Methods: Rat models of hyperoxia-induced neovascularization were established. Models were divided into blank, small-dose puerarin, and large-dose puerarin groups (n=4, each). We observed the leakage area(using fluorescein fundus angiography,FFA) ,detected the expression of PECAM-1 by immunofluorescence staining and VEGF in vitreum by ELISA. Results: Puerarin administration reduced neovascularization leakage area ,inhibited PECAM-1,VEGF expression in a dose-dependent manner. Compared with blank, The expression of VEGF was decreased (p<0.001) in the small-dose group, and the degree of reduction was significantly increased in the large-dose group (p<0.001). Conclusion: Puerarin can inhibit the hyperoxia-induced retinal neovascularization in rats by inhibiting the expression of VEGF and PECAM-1. |
